C12E8 est un tensuiactif non-ionique, son nom complet est tensioactif polyoxyéthylène glycol, il appartient à la famille des tensioactifs polyoxyéthylène éther [1]. C12E8 régule la courbure et la transition morphologique de la membrane des vésicules phospholipidiques de manière concentration-dépendante, favorisant les changements morphologiques naturels à faible concentration et induisant une déformation anormale et finalement entraînant la désintégration des vésicules à haute concentration[2-3].
Dans les cellules DC3F, C12E8 (0.05mg/mL; 45min) diminue la tension requise pour l'électroporation irréversible dans les bicouches lipidiques planes [4]. Dans les érythrocytes, de grandes invaginations plates/endovésicules avec une région centrale comprimée et une périphérie globulaire ont été trouvée dans les sections d'érythrocytes traités avec C12E8 (44μM; 60 min) [5].

Méthode de préparation | Les cellules en suspension ont été incubées pendant 45 minutes à 37°C; 0.05mg/mL C12E8 a été ajouté aux cellules traitées, tandis que le milieu dans lequel C12E8 a été dissous a été ajouté aux cellules non-traitées. La fluidité membranaire a été mesurée par la méthode de résonance paramagnétique électronique (RPE), en utilisant la sonde de spin méthyleste de 5-doxyl palmitate (MeFASL), qui est lipophile et s'incorpore donc principalement dans la bicouche lipidique membranaire. Soixante millilitres d'une solution de 0.1mM MeFASL dans l'éthanol ont été placés dans le tube en verre, puis l'éthanol a été évaporé au rotavapor. 1mL de suspension cellulaire contenant 20 × 106 cellules a été ajouté au film de MeFASL formé sur la paroi du tube en verre, et incubé pendant 15 minutes sous agitation. Après cela, la suspension cellulaire a été centrifugée et le culot a été placé dans une capillaire en verre pour les mesures RPE. A partir des spectres RPE à 4°C, la constante de désintégration hyperfine maximale 2AII , qui reflète le paramètre d'ordre, a été mesurée. |
