Oleanolic acid est un triterpénoïde pentacyclique naturel reconnu pour son efficacité dans le traitement de l'hépatite. De plus, il démontre également des effets antitumoraux sur divers types de cancers[1-3].
A des concentrations de 50 μM, 65 μM et 80 μM pendant 24 ou 28 heures, Oleanolic acid a renforcé la capacité du regorafénib à inhiber la croissance des cellules PLC, particulièrement aux doses de 65 μM ou 80 μM[4]. Administré à des concentrations de 2 μM, 4 μM et 8 μM pendant 48 heures, Oleanolic acid agit comme un activateur de Nrf2, montrant une atténuation dose-dépendante de la transition épithélio-mésenchymateuse (EMT)induite par GF-β1 dans les cellules NRK-52E[5]. De plus, oleanolic acid à une concentration de 50 μM pendant 6 heures induit la libération de prostacycline dans les cellules musculaires lisses coronaires humaines (SMC) via un mécanisme dépendant de Cox-2[6].
Oleanolic acid(50 mg/kg/j; i.g; 2 semaines) exerce un effet anti-EMT in vivo et potentialise l'effet antitumoral du regorafénib[4]. Oleanolic acid(10-100 mg/kg; ip) a démontré un potentiel anti-épileptogène significatif dans lesc crises induites par le pentylènetétrasole[7].

Méthode de préparation | Les cellules PLC (5 × 103 cellules/mL) ont été ensemencées dans des plaques de culture à 96 puits et incubées pendant 20 heures. Par la suite, les cellules ont été traitées soit avec le solvant seul, soit avec oleanolic acid (aux concentrations de 50 μM, 65 μM et 80 μM), du regorafénib, ou une combinaison d'oleanolic acid et de regorafénib. Les cellules ont ensuite été incubées pendant 24 ou 48 heures. |

Méthode de préparation | 1 × 106 cellules PLC, en suspension dans PBS, ont été injectées par voie sous-cutanée à des souris, qui ont ensuite été réparties aléatoirement en quatre groupes. Deux semaines après l'inoculaiton, les groupes expérimentaux ont reçu par administration orale soit de l'oleanolic acid (50 mg/kg/jour), soit du regorafénib (10 mg/kg/jour), soit une combinaison d'oleanolic acid (50 mg/kg/jour) et de regorafénib (10 mg/kg/jour) pendant deux semaines supplémentaires. Le groupe témoin a reçu un volume équivalent de solution saline pendant la même durée. |
