Description of PHYSALIEN

PHYSALIEN est un caroténoïde lipophile[1-2]. PHYSALIEN possède les propriétés antioxydantes et anti-inflammatoires, qui expliquent ses effets hépatoprotecteur atténue le stress oxydatif, réduit la fibrose hépatique. On l'utilise donc dans les recherches sur les maladies hépatiques et les troubles oculaires[3-4].

Les hépatocytes normaux de rat BRL-3A ont été pré-traités in vitro avec PHYSALIEN (1µM) pendant 2h, avant de les co-incuber avec de l'éthanol pendant 24h. Ce traitement a restauré l'expression des protéines clés de l'autophagie (Atg5, Beclin-1, et LC3A/B) qui avait été inhibée par l'éthanol, a réduit l'accumulation du substrat d'autophagie p62. Le pré-traitement avec PHYSALIEN a ciblé les récepteurs membranaires P2X7 et AdipoR1, ce qui a modulé les voies qui transmettrent les signaux AMPK/FoxO3a et PI3K/Akt. Cela a permis de restaurer la mitophagie inhibée par l'éthanol et d'atténuer l'activation de l'inflammasome NLRP3[5].

In vivo, sur un modèle murin de stéatose hépatique alcoolique (AFLD) chez les rats mâles C57BL/6, on a administré PHYSALIEN (25mg/kg/jour) de la 5ème à la 10ème semaine de l'expérience. PHYSALIEN a significativement amélioré les symptômes typiques de AFLD induits par la consommation d'éthanol : Amélioration de l'histologie hépatique (réduction du dépôt de gouttelettes lipidiques et des foyers inflammatoires); Baisse de taux sérique d'alanine aminotransférase (ALT); Atténuation du stress oxydatif et de l'inflammation hépatique; Cet effet hépatoprotecteur est médié par la régulation des voies qui transmettrent les signaux CYP2E1, NF-κB, et MAPK (p38 et ERK)[6]. Dans un modèle de dégénérescence photoréceptrice induite par la N-méthyl-N-nitrosourée (MNU) chez les rats mâles adultes C57BL/6J, on a administré PHYSALIEN (approximativement 0.27mg/kg), en commençant une semaine avant l'injection de MNU et pendant deux semaines au total. PHYSALIEN a considérablement amélioré la durée de vie des photorécepteurs, renforcé les réponses électrophysiologiques rétiniennes à la lumière, augmenté l'acuité visuelle des rats lésés[7].

Lignée cellulaire d'hépatocytes normaux de rat BRL-3A

Méthode de préparation

On a cultivé les cellules BRL-3A dans le milieu DMEM additionné de 10% (v/v) de sérum foetal bovin, à 37°C, 5% CO₂. On a pré-traité les cellules avec PHYSALIEN (1μM) pendant 2h, avant d'ajouter 250mM d'éthanol pour 24h.

Conditions de réaction

1µM; prétraitement de 2h

Domaines d'application

PHYSALIEN a nettement neutralisé la dérégulation induite par l'éthanol des protéines clés de l'autophagie (Atg5, beclin-1, LC3A/B) et la suraccumulation du substrat d'autophagie p62 dans les cellules BRL-3A. PHYSALIEN a aussi partiellement inversé la mitophagie inhibée par l'éthanol, ce qui se voit par une restauration de la morphologie mitochondriale (diminution du gonflement et des crêtes altérées) et une augmentation du pourcentage de mitochondries intactes. En outre, PHYSALIEN a atténué l'activation de l'inflammasome NLRP3 déclenchée par l'éthanol, réduisant l'expression de la caspase-1 clivée et la sécrétion d'IL-1β et d'IL-18.

Modèles animaux

Rats féminins Sprague-Dawley (modèle de stéatose hépatique alcoolique)

Méthode de préparation

On a administré de l'éthanol par voie intragastrique (4,0 g/kg) aux rats pendant 10 semaines. PHYSALIEN a été administré par voie orale (25mg/kg/jour) de la 5ème à la 10ème semaine. Les rats ont été sacrifiés 8h après la dernière administration.

Forme de dosage

25mg/kg/day; gavage oral; administration quotidienne pendant 5 semaines.

Domaines d'application

Le traitement avec PHYSALIEN a nettement atténué les symptômes de la stéatose hépatique alcoolique (AFLD), notamment : réduction de la perte de poids corporel; diminution du dépôt de gouttelettes lipidiques dans le foie; baisse de l'infiltration de cellules inflammatoires. PHYSALIEN a amélioré la fonction hépatique en faisant baisser le taux sérique d'ALT et de triglycérides (TG). PHYSALIEN a atténué le stress oxydatif hépatique (en réduisant l'expression de CYP2E1, MDA et 8-isoprostane, et en restaurant CAT et SOD1), a diminué l'inflammation (en réduisant TNF-α, IL-1β, IL-6, MCP-1), a réduit l'apoptose hépatique.
