Thymalfasin (Thymosin α1) est une protéine courte, fortement chargée et intrinsèquement désordonnée, dotée d'effets immunorégulateurs[1]. Thymalfasin peut augmenter l'expression des antigènes du complexe majeur d'histocompatibilité (CMH) et de la β2-microglobuline à la surface des cellules, ce qui renforce l'expression d'antigènes spécifiques aux virus et inhibe la réplication virale[2]. Thymalfasin est largement utilisé dans les modèles cellulaires et animaux pour inhiber la progression tumorale et atténuer les réponses inflammatoires[3].
Le traitement in vitro avec Thymalfasin pendant 24 heures a inhibé la prolifération des cellules SNK6, sa valeur d'IC50 est 1.334μmol/ml[4]. Le traitement avec 50μg/ml de Thymalfasin aux cellules HepG2 pendant 12 heures a induit un arrêt du cycle cellulaire et inhibé la phosphorylation d'Akt au niveau de la sérine 473[5]. Un traitement avec 130μM de Thymalfasin pendant 48h a entraîné l'apoptose et des lésions à DNA dans les cellules ZR-75-1 et les cellules MCF-7[6].
In vivo, l'administration intra-péritonéale de Thymalfasin à la dose de 0,6mg/kg/jour pendant 5 jours consécutifs a amélioré la survie et réduit la charge tumorale dans les modèles de xénogreffe de cellules MC38 chez le rat recevant une chimiothérapie intra-péritonéale hyperthermique (HIPEC)[7]. Dans un modèle murin d'infection par C. albican, une injection intra-péritonéale de 200μg/kg/jour de Thymalfasin pendant 7 jours consécutifs a atténué l'inflammation gastro-intestinale induite par C. albican, augmenté le niveau d'IL-22 et réduit la croissance fongique dans le foie[8].

Méthode de préparation | Les cellules MCF-7 sont cultivées régulièrement dans un milieu Eagle modifié de Dulbecco (DMEM), supplémenté avec 10% (v/v) de sérum bovin foetal (FBS) inactivé par la chaleur, de l'insuline (0,1 unités/ml), de la L-glutamine (2mM), du pyruvate de sodium (100mg/ml), des acides aminés non-essentiels (100µM), de la streptomycine (100mg/mL) et de la pénicilline (100 unités/mL), dans une incubatrice humidifiée à 37°C sous 5% de CO2. Les cellules ont été ensemencées dans les plaques de 96 puits à une densité initiale de 5×10⁴ cellules/puit et cultivées pendant 24h. Après incubation, les cellules ont été cultivées davantage dans un milieu contenant différentes concentrations de Thymalfasin (40, 70, 100, 130, et 160μM) pendant 12, 24, 48, 72, et 96h, respectivement. A chaque point temporel indiqué (12, 24, 48, 72, et 96h), MTT a été ajouté à chaque puits pour atteindre une concentration finale de 0,5mg/mL; le tout a été ensuite dissous dans 150µl de diméthyl sulfoxyde et mesuré à une longueur d'onde de 490nm. |

Méthode de préparation | A l'aide d'un cathéter plastique de 4cm de longueur (calibre 18G), 1×108 cellules C. albicans ont été dissoutes dans 200µL de Saline physiologique, puis inoculées par voie gastrique (i.g) chez les rats C57BL/6 pour établir un modèle d'infection gastro-intestinale. Thymalfasin a été administré par voie intra-péritonéale à la dose de 200µg/kg chaque jour pendant 7 jours. Des échantillons provenant de l'estomac et du côlon des rats ont été collectés pour une analyse histopathologique. |
