Validamycin A est un fongicide agricole qui protège les cultures du champignon Rhizoctonia solani en inhibant la tréhalase, sa valeur d'IC50 est 72µM[1]. Validamycin A inhibe également la croissance d' Aspergillus flavus (valeur MIC : 1µg/mL) et est couramment utilisé pour lutter contre des maladies telles que la brûlure de la gaine du riz et la gale noire de la pomme de terre, ainsi que pour étudier l'activité de la tréhalose dans les systèmes biologiques[2,3,4,5].
Le traitement in vitro avec Validamycin A (0.125-1μg/mL), seule ou en combinaison avec l'amphotéricine B (2µg/mL) aux cellules épithéliales bronchiales humaines (BEAS-2B) pendant 24h n'a pas induit de cytotoxicité significative. Le traitement avec Validamycin A (1μg/mL) à la souche Aspergillus flavus ATCC 204304 pendant 24h a significativement réduit la capacité d'adhésion d' A. flavus[1]. Le traitement avec Validamycin A (1μM) aux cultures en flacon agité de Rhizoctonia cerealis pendant 8 jours a considérablement réduit la teneur totale en inositol du mycélium (de 62%) et diminué le niveau d'inositol libre et hydrolysé dans le surnageant de culture[6].
In vivo, l'exposition d'environ 100 oeufs de moustiques Aedes aegypti à un milieu d'élevage contenant Validamycin A (0.1, 0.2, 0.5mg/mL) dans un volume de 200 mL (les larves étant exposées continuellement jusqu'au stade adulte) a significativement retardé les processus d'éclosion, de pupaison et d'émergence, et inhibé de manière dose-dépendante la capacité de vol des adultes (100% des adultes n'étaient pas capables de voler à la concentration de 0.5mg/mL)[7]. Validamycin A (150, 300, 600, 1200mg/kg) administré par voie orale en association avec le pentylènetétrazol (PTZ) à des poissons zèbres adultes, suivie d'un enregistrement d'électroencéphalogramme (EEG) pendant 20 minutes, a réduit de manière dose-dépendante le nombre et la durée cumulative des activités EEG similaires à des crises induites par PTZ[8].

Méthode de préparation | Les cellules BEAS-2B ont été traitées avec Validamycin A (0.125, 0.5, 1μg/mL), seule ou en combinaison avec amphotericin B (2μg/mL) pendant 24h. La toxicité de Validamycin A et de amphotericin B sur les cellules BEAS-2B a été évaluée à l'aide de Kit d'essai colorimétrique de cytotoxicité de la lactate déshydrogénase (LDH) II. Après 24h, 25µl du mélange réactionnel LDH ont été ajoutés et incubés à 37°C pendant 30 minutes. Les densités optiques (DO) ont été ensuite mesurées à 450nm à l'aide d'un spectrophotomètre. |
