VT103は選択的なTEAD1タンパク質パルミトイル化の阻害剤である。VT103は、YAP/TAZ-TEAD媒介の遺伝子転写を阻害し、TEADの自己パルミトイル化を遮断し、YAP/TAZとTEADの相互作用を破壊する[1-2]。VT103は、HER2陽性乳癌、前立腺癌、トリプルネガティブ乳癌に関連する研究に使える[3-4]。
In vitro(体外)実験で、アンジオテンシン(Ang-II;100nM)で刺激した新生マウス心臓線維芽細胞(NMCF)をVT103(10μM)で48時間処理した。VT103は、α-SMAの発現におけるAng-IIによる上昇を逆転させ、コラーゲンゲル収縮と細胞の遊走を阻害した[5]。また、VT103(10μM)で、METTL14を過剰に発現するヒト冠動脈平滑筋細胞(HCASMC)を24時間処理した。VT103は、TEAD1、PDGFRB、CTGFとCyr61の発現を阻害し、収縮マーカーACTA2、CNN1とSM22αの発現を回復させ、METTL14の過剰発現により増強された細胞の生存率、増殖と遊走を軽減した[6]。
In vivo(体内)実験で、KTOR81異種移植腫瘍を持つ雌BALB/c-nuマウスに、VT103(100mg/kg)とDabrafenib(30mg/kg)を併用して、14日間、毎日経口投与した。VT103は、42日間の観察期で、マウスの体重を著しく影響せずに、腫瘍の増殖を著しく阻害し、持続的な腫瘍の退縮を維持した[7]。NCI-H226異種移植腫瘍を持つマウスに、VT103(0.3、1、3mg/kg)を45日間、毎日経口投与した。VT103は、腫瘍の増殖を著しく阻害し、3mg/kgの用量で、VT103は処理期の間に体重を著しく影響せずに腫瘍の退縮を誘発した[8]。
