PHYSALIEN은 친지질성 카로티노이드 화합물이다. PHYSALIEN은 항산화 및 항염증 특성을 나타내고 산화 스트레스를 완화하고 간 섬유화를 경감시켜서 간 보호 효과에 기여한다. PHYSALIEN은 간 질환 및 안과 질환 관련 연구에 활용된다[3-4].
체외 실험에서 BRL-3A 쥐 정상 간세포를 PHYSALIEN (1µM)으로 2시간 전처리한 후 에탄올과 24시간 공동 배양하였다. 해당 처리는 에탄올에 의해 억제된 주요 자식 관련 단백질 (Atg5, Beclin-1 및 LC3A/B)의 발현을 회복시키고 자식 기질 p62의 축적을 감소시켰다. PHYSALIEN 전처리는 세포막 수용체 P2X7 및 AdipoR1을 표적으로 해서 AMPK/FoxO3a 및 PI3K/Akt 신호전달 경로를 조절하고 에탄올에 의해 억제된 미토콘드리아 자식을 회복시키고 NLRP3 인플라마좀의 활성화를 약화시켰다[5].
체내 실험에서 수컷 C57BL/6 쥐를 이용한 알코올성 지방간 질환 (AFLD) 모델에서 실험 5주차부터 10주차까지 PHYSALIEN (25mg/kg/일)을 경구 투여하였다. PHYSALIEN은 에탄올 섭취에 의해 유발된 전형적인 AFLD 증상을 현저하게 완화하였다. 이는 간 조직병리학 개선(지방방울 침착 및 염증 병소 감소), 혈청 알라닌 아미노전이효소(ALT) 수준 저하, 간 산화 스트레스 및 염증 완화를 포함하였다. 간 보호 효과는 CYP2E1, NF-κB 및 MAPK (p38 및 ERK) 신호 전달 경로의 조절을 통해 매개되었다[6]. 성체 수컷 C57BL/6J 쥐에서 N-메틸-N-니트로소우레아(MNU)로 유도된 광수용체 퇴행성 변화 모델에서 손상 1주 전부터 시작해서 총 2주간 매일 PHYSALIEN(약 0.27mg/kg)을 경구 투여하였다. PHYSALIEN은 손상된 쥐에서 광수용체 생존율을 현저하게 개선하고 빛에 대한 망막 전기생리학적 반응을 강화해서 시각 예민도를 증가시켰다[7].
