Description of Baicalein

Baicalein (5,6,7-trihidroxyflavona) es un compuesto flavonoide con múltiples actividades biológicas. Es un inhibidor de la xantina oxidasa con un valor de IC50 de 3.12μM[1]. Baicalein interactúa con el sitio de unión de las benzodiazepinas del receptor GABAA en preparaciones de membranas corticales de ratón con un valor de Ki de 13.1 μM[2]. Baicalein presenta actividades antitumorales y antiinflamatorias, y también puede desempeñar un papel neuroprotector y protector mitocondrial[3].

In vitro, el tratamiento con Baicalein (25 μM) durante 2 h en linfocitos esplénicos de ratón inhibió significativamente la proliferación de células T inducida por mitógenos y la secreción de citoquinas, así como la sobreexpresión de CD69 y CD25 (IL2Rα), pero no inhibió la expresión de CD54 (ICAM-1)[4]. El tratamiento con Baicalein (0-160 μM) durante 24 h en dos líneas celulares de cáncer de ovario (OVCAR-3 y CP-70) y una línea celular normal de ovario (IOSE-364) inhibió selectivamente la viabilidad de las células cancerosas, mientras que el efecto inhibidor sobre las células normales fue significativamente menor. También inhibió la expresión de oncogenes como VEGF, HIF-1α, cMyc y NFκB[5].

In vivo, Baicalein (1, 3 mg/kg) fue administrado mediante inyección intratumoral en ratones inoculados con células tumorales H-460 durante 4 semanas, lo que inhibió significativamente el crecimiento tumoral, redujo la expresión de 12-LOX y VEGF en el tejido tumoral, y disminuyó el índice de células mitóticas y la densidad microvascular[6]. Baicalein (30, 100 mg/kg) fue utilizado para tratar ratones con úlceras gástricas agudas inducidas por etanol acidificado, inhibiendo significativamente la formación de úlceras agudas, reduciendo el proceso inflamatorio, estimulando los mecanismos antioxidantes celulares e incrementando la secreción de mucosa gástrica[7].

Método de preparación

Los linfocitos esplénicos murinos teñidos con CFSE fueron pretratados con 5 a 25 μM de Baicalein durante 2 h y estimulados con Con A (mitógeno de células T) o anti-CD3/CD28 mAb. La proliferación se evaluó mediante la dilución del tinte CFSE utilizando un citómetro de flujo.

Método de preparación

Las células H-460 se administraron por vía subcutánea en el flanco dorsal izquierdo de ratones machos desnudos de 6 semanas (BALBc). Cuando el tamaño del tumor alcanzó aproximadamente 50 mm³, los animales fueron aleatorizados en grupos de control y tratamiento. Los ratones recibieron Baicalein (1 mg/kg o 3 mg/kg en 50 μl de DMSO/PBS), o un volumen equivalente del vehículo de control, mediante inyección intratumoral. La inyección intratumoral se realizó dos veces por semana, y el tamaño del tumor se midió cada 48 h utilizando un calibrador digital.

Áreas de aplicación

Los tumores H-460 tratados con Baicalein mostraron un crecimiento significativamente reducido, disminución de la expresión de 12-LOX y VEGF, así como una reducción significativa en el índice de células mitóticas y la densidad de microvasos.
