BQ-788 es un antagonista selectivo del receptor de endotelina tipo B (ETB) no peptídico con un IC50 de1.2nM[1]. BQ-788 puede unirse específicamente a los receptores ETB, bloqueando la interacción entre la endotelina y sus recepotre, inhibiendo así los efectos mediados por la endotelina, como la vasoconstricción y la proliferación celular [2]. BQ-788 se utiliza para investigar los macanismos de la ETB en el sistema cardiovascular, incluidas enfermedades como la hipertensión y la insuficiencia cardíaca[3]. BQ-788 también se puede utilizar para estudiar el papel de la ETB en el sistema nervioso, como la neuroprotección y la neuroinflamación [4].
In vitro, BQ-788 (1μM) cotratado con fibrinógeno (Fg; 4mg/mL) en células endoteliales microvasculares del corazón de rata (RHMECs) durante 45 minutos atenuó significativamente la fosofrilación de la quinasa 1/2 regulada por señal extracelular (ERK-1/2) inducida por Fg[5]. La células de la mucosa olfativa de rata pretratadas con BQ-788 (10μM) pdurante 1 semana no tuvieron un efecto significativo en la proliferación celular, pero aumentaron significativamente el nivel de expresión de la endotelina-1 endógena (ET-1)[6].
In vivo, BQ-788 (0.2, 1 y 5mg/kg) se inyectó por vía intravenosa en ratones ICR , seguido de estimulación con ET-1 (0.1μmol/kg), y BQ-788 inhibió significativamente la broncoconstricción inducida por ET-1 en ratones[7]. BQ-788 (30nmol) co-inyectado subcutáneo con ET-1 (100pmol) en ratones C57BL/6J mejoró significativamente el comportamiento de rascado inducido por ET-1[8].
