Iristectorin B es una isoflavona natural extraída de Iris tectorum y posee propiedades anticancerígenas[1]. Puede generar efectos antiinflamatorios, ya que frena la síntesis de NO provocada por el lipopolisacárido (LPS)[2]. Por cierto, se usa mucho como sustancia de referencia interna para crear métodos de extracción asistida por ultrasonidos con líquidos iónicos, pensados para aislar compuestos afines[3].
En ensayos in vitro, al tratar las células PC12 durante 24 horas con Iristectorin B (25µM), mejora su supervivencia tras el daño por privación de glucosa-oxígeno seguido de reoxigenación (OGD/R). Además, baja la apoptosis celular y logra reducir de forma marcada los niveles de expresión de TFR1, HMOX1 y SLC3A2[4].

Método de preparación |
Las células PC12 se cultivaron en un medio DMEM con alto contenido de glucosa suplementado con un 10% de suero bovino fetal (FBS) y un 1% de Penicillin/Streptomycin a 37℃ en presencia de 5% de CO2. Se agregaron 100μl de solución EBSS sin azúcar que contenía 10mM de Na2S2O4 y se incubaron durante 2h. Las células fueron reemplazadas por un medio normal para la reoxigenación y continuaron siendo cultivadas durante 24 horas, constituyendo un modelo de lesión OGD/R inducida para simular una lesión cerebral isquémica. OGD/R se complementó con medio de cultivo normal que contenía Iristectorin B 6.25µM, 12.5µM y 25µM durante 24h. Se analizó la viabilidad celular. |
