CPYPP band reversibel an die DOCK2 DHR-2-Domäne (DOCK2DHR-2) und hemmte deren katalytische Aktivität[1].
CPYPP hemmte die Guaninnukleotid-Austauschfaktor (GEF)-Aktivität von DOCK2DHR-2 für Rac1 in einer dosisabhängigen Weise mit einer halbmaximalen inhibitorischen Konzentration (IC50) von 22,8 ± 2,4 µM. Diese inhibitorische Aktivität war unabhängig von der Vorinkubationszeit, die zwischen 2 Minuten und 30 Minuten lag. Darüber hinaus zeigte CPYPP keine Toxizität, wenn es 3 Stunden bzw. 3 Tage lang in einer Konzentration von 100 µM auf Milzzellen oder Thymomzellen (BW5147α-β-) angewendet wurde[1]. Die Vorbehandlung menschlicher Neutrophilen mit CPYPP, einem niedermolekularen Inhibitor der DHR-2-Domäne von DOCK-Proteinen und damit der Rac-GEF-Aktivität von humanem DOCK2 und DOCK5, beeinträchtigte die Chemotaxis der Neutrophilen und die Produktion reaktiver Sauerstoffspezies(ROS) signifikant[2].
CPYPP reduzierte wirksam die Sekretion und Genexpression von TNF-α, IL-1β und IL-6 in der Lunge, was auf eine Wirkung von DOCK2 auf endotoxämieinduzierte Entzündungsreaktionen bei Mäusen hindeutet. CPYPP hemmte deutlich die Infiltration von Gesamtzellen, Makrophagen und Neutrophilen in die bronchoalveoläre Lavageflüssigkeit[3]. Eine Studie ergab, dass die kombinierte Verabreichung von CPYPP und C25-140 im Vergleich zur Monotherapie zu bessern Ergebnissen bei der Bekämpfung von hepatischen Ischämir-Reperfusionsschäden führte[4].

ALI wurde durch intraperitoneale (i.p.) Injektion von LPS (10 mg/kg) Körpergewicht induziert; Die Mäuse in den Gruppen LPS + Vehikel und LPS + CPYPP erhielten 10 Minuten nach der LPS-Verabreichung CPYPP (250 mg/kg) oder ein äquivalentes Volumen Vehikel per i.p.-Injektion.
