Preparation Method |
4 µg/µL polybrene was incubated at 37℃ for 15 min, and intracerebroventricularly (ICV) injection of 5 µL mixture was performed for 20 min. Rotating Rod test will take place in three days. |

Polybrene, ein quaternäres Ammoniumsalz, wurde zunächst in der klinischen Praxis als Heparin-Neutralisator eingeführt[1]. Darüber hinaus wurde Polybrene in niedrigeren Dosen häufig verwendet, um die Effizienz der virusvermittelten Genübertragung sowohl in vivo als auch in vitro zu fördern[2-5]. Polybrene fördert die Bindung von Viren an der Zelloberfläche durch Neutralisierung der elektrostatischen Abstoßung zwischen den gegenüberliegenden Bilayern[6-7].
Polybrene (0-32 µg/mL; 24h) zeigt konzentrationsabhängige Effekte. Bereits bei Konzentrationen von nur 4 µg/mL bilden sich deutliche neuritische Perlen, während eine Konzentration von 8 µg/mL zur Fragmentierung der Neuriten führt[8]. Polyethylen (8µg/mL) reduziert signifikant die Proliferationsrate mesenchymaler Stammzellen (MSCs)[9]. Polybrene verbessert die Effizienz der Lipofektion erheblich und kompensiert auch die serumvermittelte Hemmung der Lipofektion in einer Vielzahl von Zelllinien[10].
Die Injektion von Polybrene (4 µg/µL; ICV) führte bei Mäusen am dritten Tag zu motorischen Störungen. Polybrene verursacht neuronales Sterben und Gliose bei Mäusen[8].
