Beschreibung (Description)

Givosiran sodium is a subcutaneously administered siRNA targeting hepatic ALAS1 mRNA [1]. Givosiran sodium is conjugated to a trivalent N-acetylgalactosamine ligand, which can be specifically delivered to hepatocytes, resulting in downregulation of ALAS1 mRNA and preventing accumulation of neurotoxic δ-aminolevulinic acid and porphobilinogen (PBG) levels that are associated with acute porphyria attacks [2]. Givosiran sodium has been widely used to rapidly and sustainably reduce ALAS1 mRNA, decrease heme precursor levels, and lower the rate of acute attacks [3].

In vivo, Givosiran sodium treatment via subcutaneous injection at a dose of 3mg/kg, twice a week for 3 weeks, significantly reduced the liver ALAS1 mRNA level and the accumulation of 5-aminolevulinic acid (ALA) and PBG in the urine of the acute intermittent porphyria (AIP) mouse model [4].

Protokoll (Protocol)

Animal experiment [1]:

Animal models

Male AIP mice

Preparation Method

Male AIP mice (40-120 days old) were maintained under a 12h light and 12h dark cycle at a temperature of 25°C and humidity of 50%±10%, free with water and an ad libitum diet. 3,5-diethoxycarbonyl-1,4-dihydro-2,4,6-trimethylpyridine (DDC) (20mg/kg/day) and phenobarbital (PB) (90, 100, 110, and 90mg/kg/day) were administered on days 0, 1, 2, and 3, and mice were treated with subcutaneous Givosiran sodium or PBS was administered prophylactically, twice weekly for 3 weeks at a dose of 3mg/kg. A subset of mice was subsequently perfused with saline, and the livers were harvested for analysis.

Dosage form

3mg/kg; twice weekly for 3 weeks; s.c.

Applications

Givosiran sodium treatment reduced the liver ALAS1 mRNA level in AIP mice.
