ML351 ist ein selektiver 15-lipoxygenase-1 (15-LOX-1; 12/15-LOX) Inhibitor mit einer IC50 von 200nM[1]. 15-LOX-1 gehört zur Familie der Lipoxidasen und kann eine Phospholipidperoxidation in der Plasmamembran auslösen. Durch Hemmung von 15-LOX-1 kann ML351 die Produktion schädlicher oxidativer Produkte in Zellen verringern[2].
In vitro konnte die Behandlung von HT-22-Zellen mit ML351 (0-10μM) die glutamatinduzierte Zelltod dosisabhängig umkehren [1]. Eine Behandlung von Peritoneal-Makrophagen (PMφ)-Zellen für 2 Stunden mit 15-LOX-1 und der induzierten Nitrogenmonoxid-Synthase(iNOS )[3].
In vivo reduzierte eine Behandlung mit ML351 (50 mg/kg) bei Mäusen mit akuter Herzinsuffizienz nach subkutaner Injektion am ersten und fünften Tag deutlich die Myokardiinfarktgröße, erhöhte die Expression von CD11b und verminderte entzündliche Reaktionen im Herzen und Speicheldrüse [3]. Bei nicht-adipösen diabetischen (NOD)-Mäusen reduzierte eine intraperitoneale Injektion von ML351 (24 mg/kg) über 8 Wochen den Blutzucker, verringerte Marker für oxidativen Stress in pankreatischen β-Zellen und erhöhte die Aktivierungsgrade der Antioxidantienenzyme [4]. Bei Ratten, die eine orthodontische Behandlung für 14 Tage durchmachten, unterdrückte eine intraperitoneale Injektion von ML351 (50μM/kg) eine starke orthodontisch induzierte Wurzelfräsion (OIRR) und unterdrückte das Auftreten von Osteoklasen und Odontoklasen [5].

Vorbereitungsmethode | PMϕ wurden nach Injektion von kaltem Medium (DMEM mit 10% inaktiviert FBS, 1% Antibiotika/Antimykotik) in die Peritonealhöhle von männlichen C57BL V 6- Mäusen isoliert. Nach 4-6 Stunden bei 37°C, 5% CO2 in der Inkubator sind die Zellen gut haftend und bereit für Experimente: ML351 (10μM) wurde für 2 Stunden direkt an die Zellen gegeben. |
