Surfactin, a highly powerful biosurfactant produced by various strains of the genus Bacillus, exhibits antibacterial, antiviral, antitumor, and hemolytic activities[1]. Surfactin increases the permeability of Propionibacterium acnes’ cell wall, allowing alkaline phosphatase to leak into the extracellular environment, and prevents Staphylococcus aureus adhesion as well as biofilm formation on surfaces[2]. Surfactin has been widely used in preclinical models to inhibit the growth of various cancer cells and as a component of nanocarriers to improve the efficiency and targeting of drug delivery[3].
In vitro, Surfactin inhibited the growth of MCF-7 cells in a dose- and time-dependent manner, with IC50 values at 24, 48, and 72h of 82.6μM, 27.3μM, and 14.8μM, respectively[4]. Pretreatment with 60µg/ml Surfactin for 1 hour downregulated lipopolysaccharide (LPS)-induced TLR4 protein expression in macrophages and reduced the levels of IFN-γ, IL-6, iNOS, and nitric oxide (NO)[5]. Treatment with 30µM Surfactin for 24 hours induced apoptosis in LoVo cells, accompanied by DNA fragmentation, changes in membrane polarity, and an increase in the cleavage level of caspase-3[6].
In vivo, Surfactin treatment via daily intragastric administration at a dose of 80mg/kg for 6 consecutive weeks alleviated dextran sulfate sodium (DSS)-induced colitis and depression-like behaviors in mice, and mitigated gut microbiota dysbiosis and colonic intestinal barrier dysfunction[7]. Daily oral administration of Surfactin at a dose of 80mg/kg for 12 consecutive weeks improved pancreatic function and glucose metabolism, promoted insulin secretion, and inhibited glucagon release in type 2 diabetic mice[8].

Preparation Method |
MCF-7 cells were cultured in RPMI-1640 medium containing 10% fetal bovine serum, 100U/ml penicillin, 100U/ml streptomycin, and 1% glutamine in a humidified cell incubator with an atmosphere of 5% CO2 at 37°C. 1×105 cells/well were plated in 96-well culture plates. After overnight incubation, the cells were treated with varying concentrations of Surfactin (0, 10, 20, 40, 60, 80, 100μM) for 48h, and cell viability was measured. |

Male Kunming mice |
|
Preparation Method |
Male Kunming mice (4 weeks old) were housed under constant conditions (temperature 25°C; 12h light-dark cycle) in an aseptic environment and fed a basic diet and water for 7 days to allow acclimatization. All mice were fed and drank ad libitum. After being weighed, the mice were randomly divided into 3 groups (n=8 per group): Control, mice treated with orally administered 0.9% saline and treated with distilled water; Model, mice treated with orally administered saline and induced with 2% DSS solution; Surfactin, mice treated with orally administered 80mg/kg/day Surfactin and induced with 2% DSS. The colonic tissues in mice were collected 6 weeks later for analysis. |
