Description of Adenosine

Adenosine est une substance neurorégulatrice endogène contenant de l'adénine, qui est liée à la molécule de ribose (furanose) par une liaison β-N9-glycosidique[1]. Adenosine affecte presque tous les aspects de la physiologie cellulaire (comprendre l'activité neuronale, la fonction vasculaire, l'agrégation plaquettaire et la régulation hématopoïétique. Adenosine peut être utilisé pour traiter la tachycardie supraventriculaire[2].

In vitro, des différentes concentrations de Adenosine (1.0-4.0mM; 12, 24 et 48 h) ont inhibé le taux de survie des cellules HepG2 de manière temporelle et dose-dépendante, augmenté significativement le niveau de mRNA et de protéines de GRP78/BiP, PERK, ATF4, CHOP, caspase-3, et cytochrome c, réduit le potentiel de membrane mitochondriale et activé le stress du réticulum endoplasmique[3]. Adenosine (0.5-4.0mM; 36 h) a considérablement augmenté l'expression de la caspase-3 dans les cellules EC109 de manière dose-dépendante, induit l'apoptose cellulaire et l'activation de NF-κB [4].

In vivo, Adenosine (1-10mg/kg/jour; i.p.) a été administré en dose unique à des rats dans le test de nage forcée (FST) et le test de suspension de la queue (TST), racourcissant significativement le temps d'immobilité dans FST et produisant un effet antidépressif similaire dans TST[5]. Adenosine (5mg/kg/jour; i.p. six fois, à des intervalles d'une heure) a augmenté le niveau d'expression du marqueur des astrocytes réactifs inflammatoires (Lcn 2) et a simultanément amélioré l'expression de plusieurs autres gènes de cytokines pro-inflammatoires/facteurs chémotactiques de cytokines, comme Ccl 2, Cxcl 1, Ilia et Tnf [6].

Lignée cellulaire d'hépatoblastome humain HepG2

Méthode de préparation

Les cellules HepG2 ont été ensemencées dans une plaque de 96 puits (5×103 cellules/puits) dans une atmosphère humidifiée avec 5% CO2 à 37°C et traitées avec Adenosine alone (0, 1.0, 2.0, 3.0 et 4.0mM) pendant 12, 24 et 48 h; ou 10µM LY 294002 seul ou 2.0mM Adenosine en combinaison avec 10µM LY 294002 pendant 12, 24, 36 et 48h. Par la suite, 10µl MTT (5mg/ml) a été ajouté à chaque puits et les cellules ont été incubées pendant 4h supplémentaires. Après élimination du surnageant, DMSO (100µl/puits) a été ajouté pour dissoudre les cristaux de formazan bleu convertis à partir de MTT par les cellules HepG2. La viabilité cellulaire a été évaluée à l'aide d'un lecteur de microplaques à une densité optique de 560nm (Wellscan K3; KHB Labsystems, Helsinki, Finland). L'expérience a été répétée trois fois.

Rats Swiss

Méthode de préparation

Les rats Swiss de sexe indifférent, pesant 30–40g, ont été maintenus à la température ambiante constante (22–27°C) avec un accès libre à l'eau et à la nourriture, sous un cycle lumière/obscurité de 12:12 h (lumières allumées à 07:00h). Toutes les manipulations ont été effectuées entre 09:00 et 17:00h, chaque animal n'étant utilisé qu'une seule fois. Adenosine et la caféine ont été dissoutes dans une saline. Les médicaments ont été administrés par voie intra-péritonéale (i.p.) à la dose de 10mg/kg de poids corporel. Adenosine ou le véhicule a été administré par voie intra-péritonéale 30 minutes avant le test de nage forcée (FST), le test de suspension de la queue (TST) ou le test de champ ouvert. Alternativement, afin d'évaluer son effet central, Adenosine a été administré par voie intracérébroventriculaire (i.c.v.) 15 minutes avant FST ou le test de champ ouvert. Les injections i.c.v. ont été effectuées sous anesthésie à l'éther léger. De la saline ou Adenosine a été injecté en volume de 5µl, administré sur 30s, et la canule est restée en place pendant 30 sem
