Description of ChX710

ChX710 est un composé de petite taille qui prépare la réponse interféron de type I à l'ADN cytosolique. ChX710 induit l'élément de réponse stimulé par l'interféron (ISRE), des gènes stimulés par l'interféron (ISG) spécifiques, ainsi que la phosphorylation du facteur de régulation de l'interféron (IRF)3, ce qui prépare efficacement la réponse cellulaire à la transfection d'ADN par la voie STING [1].

In vitro, ChX710 (6–50μM) a été appliqué à des lignées cellulaires, notamment les cellules HEK-293 pendant 8–48 heures. ChX710 active spécifiquement le promoteur ISRE et induit la phosphorylation d'IRF3. ChX710 améliore efficacement la réponse interféron de type I cellulaire à l'ADN cytosolique (transfecté), cela entraîne une augmentation de la sécrétion d' IFN-β. À plus forte concentration (25–50µM), ChX710 présente une cytotoxicité significative qui réduit les taux d'ATP et la viabilité cellulaire.[1].

Lignées cellulaires

HEK-293T (cellules épithéliales rénales embryonnaires humaines), A549 (cellules d'adénocarcinome épithélial pulmonaire humain), MRC5 (fibroblastes pulmonaires humains), Vero (cellules épithéliales rénales de singe vert d'Afrique),PBMC primaires humaines (cellules mononucléées du sang périphérique)

Méthode de préparation

Des lignées cellulaires rapporteuses (par exemple, STING-37, une lignée HEK-293 qui exprime de manière stable la luciférase sous le contrôle d'ISRE) ou d'autres types cellulaires ont été cultivées dans du milieu Eagle modifié par Dulbecco(DMEM) supplementé avec 10% de sérum de veau foetal (SVF), de la pénicilline et de la streptomycine à 37°C et sous 5% de CO₂. Les cellules ont été traitées avec ChX710 (6-50µM) ou des composés apparentés pendant des durées définies. Pour les expériences de synergie, les cellules ont été co-traitées avec  ChX710 et transfectées avec l'ADN plasmidique ou stimulées avec  2',3'-cGAMP en utilisant le réactif de transfection JetPrime.

Domaines d'application

ChX710 active spécifiquement le promoteur de l'élément de réponse stimulé par l'interféron (ISRE). Cette activation dépend de la protéine adaptatrice MAVS et du facteur de transcription IRF1, mais elle est indépendante de STAT1/STAT2.ChX710 induit la phosphorylation d'IRF3, mais cette induction est insuffisante pour déclencher à elle seule la sécrétion d'interféron de type I (IFN-α/β). ChX710 induit un profil modeste et spécifique du type cellulaire de gènes stimulés par l'interféron (ISG). De manière cruciale, ChX710 prépare efficacement et protentialise de façon synergique la réponse interféron de type I cellulaire à l'ADN cytosolique (transfecté), ce qui entraîne une augmentation forte et dépendante des ISG. ChX710 présente une cytotoxicité signficative à des concentrations plus élevées, réduisant les niveaux d'ATP cellulaire et la viabilité cellulaire
