Description of γ-Aminobutyric acid (GABA)

GABA est un neurotransmetteur inhibiteur endogène essentiel dans le cerveau des mammifères, ses valeurs d'EC50 sont respectivement 0.17, 2.1, 2.1, et 1.4μM pour α6, α1, α4, et α5 du récepteur GABAA[1]. GABA est chargé dans les vésicules synaptiques via le transporteur vésiculaire des acides aminés inhibiteurs et module l'excitabilité neuronale pour maintenir des fonctions cérébrales équilibrées[2]. GABA a été largement utilisé dans différents modèles animaux pour réguler les réponses immunitaires, interférer avec la croissance tumorale et atténuer l'anxiété[3].

Le traitement in vitro avec GABA (100µM) pendant 72 heures a significativement inhibé la prolifération des cellules SW480, SW620 et HCT116 et induit un arrêt du cycle cellulaire[4]. Le traitement avec 100µM de GABA aux cellules B des îlots pancréatiques de rat pendant 4 jours a favorisé la prolifération cellulaire, entraînant une augmentation de l'expression nucléaire de p-Akt Ser473 et p-p70S6K Thr421/Ser424[5].

Le traitement in vivo avec GABA via administration orale pendant deux semaines à une dose de 100mg/kg/jour a significativement amélioré l'état métabolique des rats atteints du syndrome des ovaires polykystiques (SOPK) et réduit le poids corporel, l'indice de masse corporelle, les taux de testostérone, le niveau d'insuline et la masse grasse chez les rats diabétiques[6]. L'injection intra-péritonéale de GABA à une dose de 1,5mg/kg par jour pendant 3 mois a amélioré la glycémie et le niveau d'insuline, l'état métabolique et la masse grasse chez les rats diabétiques[7]. Une dose unique de pré-traitement par GABA (10mg/kg; i.p.) administrée 1 heure avant a significativement augmenté le taux de survie des rats atteints de lésion pulmonaire aiguë (LPA) induite par le lipopolysaccharide (LPS), atténué la lésion inflammatoire et l'oedème pulmonaire, diminué la teneur en myéloperoxydase (MPO), réduit la régulation du niveau des cytokines pro-inflammatoires IL-1β et TNF-α, et augmenté la régulation de l'expression de la cytokine anti-inflammatoire IL-10[8].

Cellules SW480

Méthode de préparation

Les cellules SW480 ont été cultivées dans un milieu d'Eagle modifié de Dulbecco (DMEM) supplémenté de 10% de sérum de veau foetal (SVF), et maintenues à 37°C dans une atmosphère d'air humidifié contenant 5% CO2. Les cellules ont été ensemencées à une densité de 1300 cellules/puits dans des plaques de 96 puits contenant 100µl de milieu DMEM. Vingt-quatre heures plus tard, les cellules ont été incubées avec GABA (0, 10, 50, et 100µM) pendant 24, 48, et 72h, respectivement. Le dosage de la prolifération cellulaire a été réalisé par le test CCK-8, et les valeurs de densité optique à 450nm ont été mesurées.

Modèles animaux

Rats mâles C57BL/6

Méthode de préparation

128 rats mâles C57BL/6 sains âgés de 6 à 8 semaines, pesant 18 à 22g, ont été élevés en cages dans un environnement exempt de pathogènes spécifiques (EPS), avec une alimentation et une hydratation adéquates, et la température du laboratoire a été maintenue autour de 25°C. 36 rats ont été prélevés et anesthésiés par injection intra-péritonéale de 1g/kg d'hydrate de chloral à 4%. Les rats ont été divisés aléatoirement dans les 3 groupes suivants, 12 rats par groupe : groupe témoin (0,2ml de sérum physiologique par injection intra-péritonéale), groupe LPS (30mg/kg de LPS par injection intra-péritonéale), groupe GABA+LPS (10mg/kg de GABA administré par injection intra-péritonéale 1 heure avant l'administration de LPS). Les rats ont été anesthésiés 6 heures après l'injection de LPS ou de sérum physiologique, et les tissus pulmonaires ont été collectés pour analyse.
