HBC (HBC 530) est un colorant synthétique de type GFP. Sa longueur d'onde maximale est de 485nm et la longueur d'onde d'émission maximale est de 530nm[1]. HBC possède un accepteur d'électrons structurellement rigide et un fort donneur d'électrons, propriété qui lui donne la capacité d’interagir avec des molécules d’ARN de différentes longueurs et d'émettre une fluorescence intense[2]. Le canal d'imagerie par fluorescence unique fourni par HBC a reçu une utilisation étendue dans la conception de plates-formes de capteurs luminométriques pour le marquage d'ARN de multiple couleurs ou l'imagerie des interactions ARN-ARN intracellulaires [3]. HBC peut être utilisé dans la détection in situ de protéases spécifiques de tumeurs et pour l'imagerie optique animale dans des expériences d'analyse in vitro [4].

Analyse de la métalloprotéinase 2 (MMP-2) basée sur HBC530
Ce protocole fournit uniquement une ligne directrice et doit être modifié selon vos besoins spécifiques.
1. Préparation de la solution de HBC530
Une solution de travail de 2μM de HBC530 est préparé à partir d'une solution mère de 1mM de HBC530 (dissoute dans DMSO). NOTE: La solution de travail doit être préparée fraîche le jour de l'expérience.
2. Imagerie de MMP-2 par HBC530 dans la matrice extracellulaire (ECM)
(1) Pour l'imagerie de MMP-2 dans l'ECM, les cellules HePG2 sont prétraitées avec TGF-β durant 24h.
(2) Un volume de 10μl d'une sonde chimérique protéine-acide nucléique (PNCP) à 100μM est d'abord préparé dans le tampon de réaction (50mM HEPES, 50mM NaCl, 10μM MnCl2 et 1mM MgCl2) à 37°C durant 6h, puis 100μl du tampon de transcription contenant 1μM de PNCP et 4mM de rNTP sont ajoutés à la boîte de culture cellulaire durant 2h.
(3) La solution de travail de HBC530 à 2μM est incubée avec les cellules HepG2 durant15min. Un lecteur de microplaque est utilisé pour enregistrer les signaux de fluorescence.
3. Mises en grade
(1) Opération à l'obscurité: La solution de travail de HBC530 a une demi-vie de 4 h à 4°C, et elle est préparée et utilisée à la demande.
(2) Durant toute l'exprérience, veuillez à porter des gants de protection pour des précautions de sécurité appropriées.
(3) Les réactifs utilisés dans cette exprériences sont à usage exclusif de l'étude scientifique.
