Description of TH588

TH588 est un inhibiteur de MTH1 (NUDT1; IC50=5nM)[1-2]. TH588 agit par inhibtion de MTH1. Il empêche les cellules cancéreuses d'éliminer les nucléotides oxydés. Ce phénomène provoque des dommages à l'ADN. Il active aussi la réponse de mort cellulaire liée à ATM-p53 et la réparation d'ADN. TH588 peut être utilisé dans les études sur l'adénocarcinome pulmonaire, le cancer du sein, le cancer du côlon et le mélanome [3-4].

In vitro, TH588 (5-30µM) a été utilisé pour traiter des ligées cellulaires comme U2OS, HeLa et RPE-1 durant 0 à 4 heures. TH588 inhibe la polymérisation de la tubuline et la dynamique des microtubules. Il réduit fortement le renouvellement des microtubules dans le fuseau mitotique. Ce changement provoque des défauts d'alignement des chromosomes. TH588 entraîne un arrêt en mitose, la mort des cellules ou une division avec chromosomes mal alignés [5]. TH588 (1–8µM) a été utilisé sur des cellules H460 de cancer du poumon humain, des cellules U2OS d'ostéosarcome, des cellules HeLa et d'autres lignées durant 0 à 48 heures. TH588 prolonge fortement la durée de la mitose. Il active la voie de surveillance mitotique USP28-p53. Cette activation provoque un arrêt du cycle cellulaire en phase G1 du cycle suivant[6].

In vivo, TH588 (50µM) et le photosensibilisateur Ce6 (20µM) ont été chargés ensemble dans le nanovecteur T&C@SCLMs. Cette préparation a été administrée par injection dans la veine caudale (0.2ml; une fois tous les 5 jours; durant 20 jours) à des souris nues avec des tumeurs solides A431. TH588 augmente fortement l'effet de la thérapie photodynamique(PDT). Il réduit le volume des tumeurs. Il augmente aussi la proportion d'apoptose des cellules tumorales[7]. TH588 (30mg/kg) a été administré par injection sous-cutanée chaque jour durant 2 à 3 semaines à des souris nues BALB/c-nu avec xénogreffes de cellules de cancer du sein humaines MCF7, MDA-MB-231 ou MDA-MB-453. TH588 réduit fortement la croissance des tumeurs sans provoquer la toxicité importante dans le sang ou le foie.[8].

Protocol of TH588

Expériences cellulaires [1]:

Lignées cellulaires

Cellules H460 de cancer du poumon humain, cellules U2OS d'ostéosarcome et cellules HeLa

Méthode de préparation

Les cellules ont été maintenues dans un milieu DMEM (milieu de Eagle modifié de Dulbecco) avec 10% de sérum foetal bovin (FBS) et des antibiotiques à 37°C, 5% de CO₂. Les cellules ont été traitées avec TH588 (1-8μM) durant 0-48 heures.

Conditions de réaction

1-8μM; 0-48 heures.

Domaines d'application

TH588 a réduit rapidement la mobilité d'extrémités positives des microtubles. Il a perturbé l'assemblage du fuseau mitotique et l'alignement des chromosomes. Il a aussi prolongé la mitose de manière dépendante de concentration. TH588 a activé la voie de surveillance mitotique USP28-p53. Cette activation a provoqué un arrêt du cycle cellulaire en phase G1 du cycle suivant. Ces effets ne dépendent pas de sa cible prévue, l'inhibtion de MTH1.
Expériences animales [2]:

Modèles animaux

Souris nues femelles BALB/c-nu avec xénogreffes de cellules de cancer du sein humaines MCF7, MDA-MB-231 ou MDA-MB-453.

Méthode de préparation

Les souris ont reçu une injection sous-cutanée de cellules cancéreuses. Quand les tumeurs ont atteint une taille visible (environ 2mm de diamètre moyen), les souris ont reçu chaque jour une injection sous-cutanée de TH588 (30mg/kg) ou du véhicule témoin durant 2 à 3 semaines. 

Forme de dosage

30mg/kg; s.c.; quotidiennement durant 2-3semaines.

Domaines d'application

Le traitement avec TH588 a fortement réduit la croissance des tumeurs. Il a provoqué une régression de plus de 90% de tumeurs MCF7 et MDA-MB-231 et il a complètement éliminé les tumeurs MDA-MB-453. TH588 a aussi limité la prise de poids des souris, mais aucune toxicité importante du sang ou du foie n'a été observée.
