Description of Inosinic acid (IMP)

Inosinic acid (IMP)은 퓨린 대사의 핵심 물질이면서 중요한 식품 감미료이다. 아데노신산(AMP) 및 구아노신산(GMP)으로 전환될 수 있고 세포 내 에너지 대사에 참여한다[2]. 식품첨가물(E630)로 일반적으로 사용되고 L-글루탐산나트륨(MSG)과 병용 시 감미 효과를 현저하게 높이고 사료 첨가제로도 활용된다[3]. 면역 조절 및 신경 보호 분야에서도 연구 가치가 기대된다[4].

체외 실험에서 15 mM Inosinic acid으로 lineage⁻/low 골수 세포를 15–30분 처리하면 pAkt 및 pS6 신호경로가 활성화되고 막상 cKit 발현이 증가하고 과립-대식세포 전구세포(GMP)의 증식과 Gr-1⁺ 과립구 생성이 촉진된다[5].

체내 실험에서 7 Gy 전신 조사를 받은 암컷 Kv:SHK 쥐에게 조사 15분 후 Inosinic acid 45 mg/kg을 복강내 투여하면 치사량의 방사선 조사 이후 생존율이 현저하게 상슉하고 방사선 유도 백혈구 감소증 및 혈소판 감소증이 완화되고 골수와 흉선 세포의 DNA 손상 수준이 낮아진다 [6]. 10–12주령 C57BL/6J 패혈증 모델 쥐에게 Inosinic acid 1000 mg/kg을 복강내 주입하면 패혈증에서 TNF-α 생성이 현저하게 억제되고 IL-10 수준이 증가한다 [7].

계통-/저 골수세포 (머린 조혈간세포)

제조 방법

야생 형 C57BL/6 쥐 골수에서 lineage cell depletion kit로 계통-/저 골수세포를 분리하였습니다. 세포는 10 ng/mL 줄기세포 인자, 10 ng/mL 혈소판 생성 호르몬, 10 ng/mL IL-3가 보충된 SFEM 배지에서 37 °C, 5 % CO₂로 배양하였고 0–15 mM Inosinic acid으로 24–72시간 처리하였습니다.

응용 분야

Inosinic acid 처리는 용량 의존적으로 과립-단핵 전구세포(GMP)의 증식과 Gr-1⁺ 과립구 생성을 현저하게 촉진하였습니다. Inosinic acid은 GMP 세포 내 BrdU 결합량을 대조 그룹 대비 1.3배 증가시켰고 Gr-1⁺ 세포 비율을 1.4배 높였습니다.

제조 방법

쥐에게 LPS(10 mg/kg, 복강내) 도전 30분 전에 Inosinic acid(1000 mg/kg, 복강내)을 투여하였습니다. LPS 투여 90분 후 혈장과 장기(비장, 간, 폐)를 채취해서 사이토카인 분석을 수행하였습니다. 세균 패혈증 모델에서는 맹장 결찰 천공(CLP) 또는 비강 내 세균 감염(폐렴사슬알균 및 대장균) 1시간 전에 Inosinic acid(1000 mg/kg, 복강내)을 전처치하였습니다.

Inosinic acid은 내독소혈증 쥐에서 TNF-α 생성을 현저하게 억제하고 IL-10 수준을 증가시켰습니다
