Itanistomig (LB-101) is a tetravalent bispecific antibody targeting PD-L1 and CD47. Itanistomig blocks PD-L1 and achieves tumor enrichment through binding to PD-L1, and also exerts conditional CD47 blocking activity via cleavage of the hinge linker in the PD-L1-positive tumor microenvironment. Itanistomig induces antibody-dependent cellular phagocytosis in human CD14 + cells and drives tumor regression. Itanistomig can be used in research related to solid tumors [1] [2].
In Vivo, Itanistomig (administered systemically at equimolar doses with Atezolizumab ) eradicates tumors in 87.5% of MC38 hPD-L1+ syngeneic mouse models, and induces durable anti-tumor immunity to prevent tumor growth upon rechallenge[1].
In Vitro, Itanistomig strongly enhances antibody-dependent cellular phagocytosis by human CD14+ cells[1].
