NFYi5 is a nuclear transcription factor-Y ( NF-Y ) inhibitor. NFYi5 disrupts the binding of NF-Y to DNA, accelerates the ubiquitin-independent degradation of the NF-YA subunit, and reduces the transcriptional activity of NF-Y. As an antimitotic agent, NFYi5 decreases the mRNA levels of NF-Y target genes without affecting the expression of housekeeping genes, and inhibits cell proliferation. NFYi5 can be used in the research of tissue fibrosis [1].
In Vitro, NFYi5 (0.01-100 μM; 18 h) inhibits NF-Y-dependent reporter gene activity in a dose-dependent manner, with an IC50 of 19.95 μM in human cardiac fibroblasts and 12.73 μM in rat cardiac fibroblasts, while exerting no effect on non-NF-Y promoter activity[1]. NFYi5 (10-20 μM; 18 h) selectively reduces the mRNA levels of NF-Y target genes in human and rat cardiac fibroblasts without affecting the expression of housekeeping genes[1]. NFYi5 (5-20 μM; 24-48 h) inhibits the proliferation of human and rat cardiac fibroblasts in a dose-dependent manner[1].
