1-Aminobenzotriazole (ABT) es un inhibidor no específico e irreversible del citocromo P450 (CYP), activo por vía oral[1]. El 1-Aminobenzotriazol es un sustrato e inhibidor de la N-acetiltransferasa (NAT)[2].
In vitro, el tratamiento con 1-Aminobenzotriazol (1mM) de hepatocitos humanos en cultivo en sándwich (SCHH) durante 0-60 minutos inhibió significativamente las actividades intracelulares de CYP3A, CYP2C9, CYP2D6 y CYP1A2. La actividad residual de CYP3A fue del 3% en comparación con el control, mientras que las actividades residuales de CYP2C9, CYP2D6 y CYP1A2 se mantuvieron entre el 5% y el 35%[3].
In vivo, la administración oral de 1-Aminobenzotriazol (0, 50, 100, 200, 300 mg/kg) a ratas SD inhibió significativamente el metabolismo hepático de MDZ, pero no pudo inhibir completamente la formación de 1'-OH MDZ[4]. La administración oral de 1-Aminobenzotriazol (50 mg/kg) a ratones machos durante 3 días suprimió el efecto letal dominante en espermatocitos inducido por acrilamida[5]. El tratamiento de ratas con 1-Aminobenzotriazol (78 mg/kg) aumentó el peso de los órganos y la mortalidad, además de aumentar la sensibilidad al fallo multiorgánico (MOF) inducido por inflamación[6].
