AICAR (también llamado acadesina) es un nucleósido de purina. Se identificaron tres aplicaciones farmacológicas de AICAR: i) estimulación en condiciones isquémicas de la producción cardíaca del vasodilatador, adenosina [1]; ii) inhibición de la gluconeogénesis hepática a nivel de fructosa-1,6-bisphosphatase [2], de potencial terápéutico en la diabetes; y iii) estimulación de la proteína quinasa activada por AMP (AMPK), aplicada inicialmente para inhibir la síntesis hepática de triglicéridos y colesterol [3].
AICAR (tratado 48 horas) inhibió la proliferación celular de las líneas celulares de linfoma celular del manto (MCL), REC-1, JEKO-1, UPN-1, JVM-2, MAVER-1 y Z-138, con IC50s de 0.28, 0.59, 0.64, 0.98, 0.50, y 0.14 Mm respectivamente [4]. AICAR inhibió el crecimiento y el agotamiento de los grupos de nucleótidos de pirimidina en fibroblastos [5], aceleró el llenado de los grupos de nucleótidos de purina en el corazón [6], la inhibición de ácidos grasos, la síntesis de esterol y la gluconeogénesis en los hepatocitos, y el aumento de la absorción de glucosa en el músculo [7].
AICAR (500 mg/kg) inyectado por vía intraperitoneal en ratones C57BL/6J 1 hora antes de la administración de LPS. El LPS indujo la expresión del mRNA de TF en muchos órganos principales, incluidos el pulmón y el hígado [8]. Una administración diaria de 400mg/kg de AICAR en ratones previamente inoculados con un xenotrasplante de MCL redujo significativamente la carga tumoral en comparación con los animales de control, tan pronto como 7 días de tratamiento [9].
