DPQ es un inhibidor selectivo y potente de la poly(ADP-ribose) polymerase-1 (PARP-1). Puede atravesar la barrera hematoencefálica y se emplea para analizar la reparación del daño del DNA y procesos celulares relacionados[1-2]. DPQ sirve para investigar los fenómenos inflamatorios y los mecanismos de degeneración celular en múltiples patologías. En tre ellas destacam la lesión pulmonar aguda, la lesión por isquemia-reperfusión miocárdica y las enfermedades neurodegenerativas. Además, presenta efectos neuroprotectores y cardioprotectores[3-4].
In vitro, el pretratamiento de células de cáncer de pulmón no microcítico (A549 y H1299) con DPQ (1μM) durante 4 horas inhibe procesos clave. Esto ocurre tras exponerlas a irradiación con iones carbono (0.5–2Gy) mientras 24 horas. Disminuye de forma la migración celular, la capacidad de cicatrización y la actividad y expresión de las metaloproteínas MMP-2 y MMP-9[5]. El pretratamiento de células HeLa con DPQ (20–150μM) durante 1 hora, seguido de estimulación con PMA (50nM) durante 3 horas, reduce la expresión génica del receptor H1 de histamina (H1R). También disminuye la actividad del promotor de H1R[6].
In vivo, el tratamiento previo de ratones C57BL/6 con DPQ (15mg/kg) 24 horas antes de la lesión renal reduce las alteraciones patológicas por isquemia-reperfusión. Dicha lesión consiste en 45 minutos de isquemia y 6 horas de reperfusión. El fármaco evita la sobreactivación de PARP-1. Alivia la necrosis tubular aguda, la vacuolización celular y la pérdida del orillo en cepillo. También reduce el desprendimiento de células epiteliales tubulares y la formación de cilindros urinarios [7]. En ratones C57BL/6 pretratados con LPS (7.5mg/kg) mientras 30 minutos, la inyección intraperitoneal de DPQ (10μg/kg) durante 6 horas presenta efectos antiinflamatorios claros. Disminuye de forma la infiltración de neutrófilos y la actividad de mieloperoxida (MPO) en tejido pulmonar. Además, reduce los niveles de mRNA de factores proinflamatorios como TNF-α, IL-1β e IL-6[8].
