Description

Brigimadlin is an orally active MDM2-p53 antagonist. Brigimadlin restores wild-type p53 function by blocking the MDM2-p53 interaction, thereby inducing tumor cell cycle arrest or apoptosis[1-2]. Brigimadlin can be used in research related to MDM2-amplified solid tumors such as biliary tract cancer, pancreatic cancer, and lung cancer[3-4].

In vitro, TP53 wild-type brain tumor stem cells were treated with Brigimadlin (1pM-100nM) for 1-5 days. Brigimadlin was able to significantly reduce cell viability and induce cell death, while restoring p53 tumor suppressor function, including inducing p21 expression and apoptosis[5]. TP53 wild-type EHEB chronic lymphocytic leukemia cells were treated with Brigimadlin (30–100nM) for 24-72 hours. Brigimadlin significantly stabilized the p53 protein, upregulated p21WAF1 expression, induced cleavage of PARP, and activated caspase-3/7[6].

In vivo, Brigimadlin (0.125-2mg/kg) was administered orally once per week to mice bearing MDM2-amplified glioblastoma patient-derived xenografts (PDX). Brigimadlin significantly delayed tumor growth and prolonged survival[7]. Brigimadlin (10mg/kg) was administered as a single intracerebroventricular injection 60 minutes after intracerebral hemorrhage modeling in C57BL/6 mice. Brigimadlin significantly improved neurological deficit scores after intracerebral hemorrhage, reduced levels of MDA, Fe²⁺, and ROS in brain tissue, and decreased cell death and pro-inflammatory factor release[8].

Protocol

Cell experiment [1]:

Cell lines

EHEB (TP53 wild-type chronic lymphocytic leukemia cell line) and MEC-1 (TP53 mutant/deleted chronic lymphocytic leukemia cell line)

Preparation Method

Cells were maintained in RPMI-1640 medium supplemented with 10% fetal bovine serum (FBS) and 100U/mL penicillin/streptomycin at 37°C in a humidified atmosphere containing 5% CO₂. Cells were treated with Brigimadlin (30–100nM) alone or in combination with Napabucasin (BBI608).

Reaction Conditions

30–100nM; 24-72h

Applications

In TP53 wild-type EHEB cells, Brigimadlin monotherapy induced marked p53 protein stabilization, upregulation of the downstream target p21WAF1, cleavage of PARP (cPARP), and activation of caspase-3/7, leading to apoptosis. The combination of Brigimadlin with the STAT3 inhibitor Napabucasin synergistically enhanced these apoptotic markers. In contrast, TP53 mutant MEC-1 cells were resistant to Brigimadlin, showing no p53 stabilization, p21 upregulation, or PARP cleavage.

Animal experiment [2]:

Animal models

Athymic nude mice and Rag1-/-Abcb1a-/-Abcg2-/-(DKO) mice bearing patient-derived xenograft (PDX) tumors (subcutaneous or orthotopic)

Preparation Method

Subcutaneous or orthotopic tumors were established in mice. Treatment with Brigimadlin (0.125-2mg/kg) was initiated when subcutaneous tumors reached approximately 250 mm³ or 14 days after orthotopic tumor implantation.

Dosage form

0.125-2mg/kg; oral gavage; weekly.

Applications

Brigimadlin monotherapy significantly delayed tumor regrowth and extended survival in mice bearing MDM2-amplified glioblastoma PDXs. The efficacy was profoundly greater in efflux transporter-deficient (DKO) mice. Brigimadlin enhanced the response to fractionated radiation therapy in combination studies.
