Apigenin, oder 4′,5,7-trihydroxyflavone, ist ein bioaktives Flavonoid, das in vielen Pflanzen vorkommt und für seine entzündungshemmenden, antioxidativen und krebshemmenden Eigenschaften bekannt ist. Apigenin ist ein kompetitiver Inhibitor von CYP2C9 mit einem Ki-Wert von 2 µM[1]. Es hemmt die Aktivierung des Wnt/β-Catenin-Signalwegs und die Aktivität von NF-κB und blockiert die Phosphorylierung von c-Met und seinen nachgeschalteten Effektoren[2].
In vitro zeigt Apigenin (6.25-100 µM) dosis- und zeitabhängige zytotoxische Effekte auf maligne Mesotheliom(MM)-Zelllinien[2]. Nach 72-stündiger Behandlung mit Apigenin waren die IC50-Werte für MM-B1, MM-F1 und H-Meso-1-Zellen 49.16 ± 2.52µM, 46.95 ± 1.69µM bzw. 34.31 ± 1.55µM[2]. Die 48-stündige Behandlung von SGC-7901-Zellen mit Apigenin-Konzentrationen von 20, 40 und 80µmol/L führte zu Apoptoseraten von 5.76%, 19.17% bzw. 29.30%. Apigenin zeigte dosisabhängige Effekte auf Wachstum, Koloniebildung und Proliferation von SGC-7901-Zellen[3]. Apigenin (0-50 µg/ml) hemmte die Proliferation von T-24-Zellen dosisabhängig und erhöhte nach 12 Stunden die ROS-Konzentration bei gleichzeitiger Depletion von GSH in T-24-Zellen[4].
In vivo reduzierte die Behandlung mit Apigenin (20mg/kg, intraperitoneal) bei C57BL/6-Mäusen das Wachstum transplantierter maligner Mesotheliom-MM-Zelllinien in der Bauchhöle der Mäuse[2]. Die Behandlung mit Apigenin in Dosen von 125mg/kg oder 250mg/kg bei Mäusen mit ADR-induzierter Kardiomyopathie über eine Magensonde über 1/ Tage erhöhte die Phosphorylierung von AKT und mTOR in Herzzellen sowie die Expression von PI3K und reduzierte Apoptose und Autophagie in Herzzellen[5].

Vorbereitungsmethode | Gruppen von 6- bis 8-wöchigen C57BL/6-Mäusen(6 Mäuse pro Gruppe) wurden eine Woche nach der Pristan-Injektion(250 µl) intraperitoneal(i.p.) mit 0.2 ml einer Suspension mit 1×106 #40a-Zellen in PBS inokuliert. Die Mäuse wurden wöchentlich i.p. mit 20mg/kg Apigenin(gelöst in DMSO und dann verdünnt in 400 µl PBS) behandelt. Die Kontrollgruppe erhielt wöchentlich die gleiche Menge Vehikellösung injiziert. |
