Aprepitant ist ein neuartiger, hochselektiver Neurokinin-1(NK-1)-Rezeptor-Antagonist[1]. Aprepitant hat einen Kd-Wert von 86 pM für den humanen NK-1-Rezeptor. Die IC50-Werte betragen 0.1 nM(Mensch), 4 nM(Ratte) und 0.7 nM(Frettchen)[2].
In vitro entfaltete Behandlung mit 5-30 μM Aprepitant über 24-48 Stunden zytotoxische und antiproliferative Effekte auf Nalm-6-Zellen in dosis- und zeitabhängiger Weise[3]. Der IC50-Wert von Aprepitant gegen HEK 293-Zellen war >90μM[2]. Aprepitant hemmte bei Konzentrationen von 10-60 mM über 72 Stunden das Zellwachstum aller Melanom-Zelllinien in konzentrationsabhängiger Weise[4].
In vivo schwächte die intraperitoneale Gabe von Aprepitant (10mg/kg/Tag) über 3 Tage die durch Psychostimulanzien(AMPH und Kokain) induzierte konditionierte Platzpräferenz(CPP) ab. Gleichzeitig verstärkte es die durch Morphin induzierte CPP[5].

Vorbereitungsmethode | Eine Stammlösung von Aprepitant mit einer Konzentration von 10 mM wurde durch Auflösen der Substanz in 0.1% DMSO hergestellt. Zellen wurden mit entsprechenden Mengen der Aprepitant-Stammlösung behandelt, um Konzentrationen von 5, 10, 15, 20 und 30 µM zu erreichen. Zusätzlich zu den 4 Negativkontrollen(ohne Inhibitor) wurden Zellen mit der entsprechenden DMSO-Konzentration als alternative Negativkontrolle behandelt. |

Anwendungen | Aprepitant verringerte die Lebensfähigkeit und die Zellzahl, erhöhte die DNA-Syntheserate, führte zu einer Ansammlung von Zellen in der G1-Phase und verringerte die Zellpopulation in der S-Phase dosis- und zeitabhängig. |
