Description of Eact

Eact est un activateur hautement spécifique et puissant de TMEM16A, qui active TMEM16A lorsque la concentration d'ions calcium augmente[1]. En absence de Ca²⁺ , Eact augmente nettement le courant Cl- , et accroît l'amplitude et la fréquence de la contraction de l'iléon isolé de cobaye [2]. Eact a été largement utilisé pour réguler la concentration du muscle lisse des artères coronaires et pour contrôler le potentiel membranaire[3].

In vivo, un traitement par Eact à dose unique (20μl; 4.67mM) par injection dans la patte d'une souris peut induire une hyperalgésie forte et persistante à la chaleur durant 60 minutes[4]. Trente minutes avant le test d'épilepsie, une dose nique d' Eact (10mg/kg) a été adminsitrée par gavage gastrique, ce qui a nettement réduit la sévérité des crises évoquées par stimulation auditive et a diminué la fréquence des crises de course sauvage(WRS) chez les rats[5].

Rats génétiquement prédisposés à l'épilepsie (GEPR-3s)

Méthode de préparation

Des rats GEPR-3s (âgés de huit semaines) ont été placés dans une salle à température et humidité contrôlées, avec un cycle lumière/obscurité de 12h/12h et un accès libre à la nourriture et à l'eau. Eact (10mg/kg) et T16Ainh-A01 (10mg/kg) ont été dissous dans de l'acide méthanesulfonique (1%) et de l'eau stérile par sonication(80kHz; puissance 100% ); les solutions ont été filtrées et adminstrées 30min avant le test de provocation des crises. Le véhicule, Eact et T16Ainh-A01 ont été administrés par voie orale par gavage gastrique. Pour induire les crises, un stimulus acoustique composé de sons purs à un niveau de pression sonore de 100 à 105 décibels a été présenté jusqu'à ce qu'une crise soit déclenchée, ou jusqu'à ce que 60s se soient écoulées sans activité de crise.
