Description

2-Phospho-L-ascorbic acid trisodium salt, a stable form of Ascorbic acid (Asc), is frequently employed as source of Asc in cell culture[1]. 2-Phospho-L-ascorbic acid trisodium salt promotes osteoblast differentiation by enhancing collagen synthesis and alkaline phosphatase (ALP) activity[2]. 2-Phospho-L-ascorbic acid trisodium salt has been widely used as a substrate for detecting target IgG and for determining hexavalent tungsten (W(VI)) in water[3-4]. 2-Phospho-L-ascorbic acid trisodium salt has been widely used in the field of regenerative medicine to induce fibroblasts to produce extracellular matrix (ECM) for the preparation of tissue-engineered matrices[5].

In vitro, 2-Phospho-L-ascorbic acid trisodium salt treatment (300μM) for 2 weeks promoted the proliferation of human corneal endothelial cells (HCECs) and reduced the level of 8-OHdG in mitochondrial DNA[6]. Treatment with 100µM 2-Phospho-L-ascorbic acid trisodium salt for 3 weeks stimulated collagen production and promoted cell proliferation in skin fibroblasts[7].

Cell experiment [1]:

Cell lines

Human forearm skin fibroblasts

Preparation Method

Human forearm skin fibroblasts (5×104 cells/well) were plated onto 35-mm dishes and grown in DMEM medium supplemented with Fungizone (250µg/L), penicillin G (50mg/L), dihydrostreptomycin (50mg/L), N-2-hydroxyethylpiperazine-N-2-ethanesulfonic acid (1g/L), and 10% fetal bovine serum (DMEM) in the absence or presence of 2-Phospho-L-ascorbic acid trisodium salt (0, 100, 200, 400, 600, 800, and 1000µM), 0.15mM magnesium chloride and 0.1mM sodium Lascorbate at 37°C for 3 weeks in an atmosphere of 5% CO2. Cell proliferation was measured.

Reaction Conditions

0, 100, 200, 400, 600, 800, and 1000µM; 3 weeks

Applications

2-Phospho-L-ascorbic acid trisodium salt treatment markedly enhanced cell proliferation of human forearm skin fibroblasts in a dose-dependent manner.
