Description of tBPC

tBPC est un modulateur allostérique positif sélectif du récepteur Y4. tBPC renforce l'activation de ce récepteur Y4 dans la voie qui transmet les signaux G-protéiques et le recrutement de l'arrestine-3. De ce fait, il potentialise les réponses du récepteur Y4 au polypeptide pancréatique, au neuropeptide Y et au peptide YY. Par ailleurs, il garde une bonne spécificité, il ne module que le récepteur Y4, ne touche pas aux récepteurs Y1, Y2 ou Y5[1].

Le traitement in vitro avec tBPC (5.1μM-30μM) aux cellules COS7 et HEK293 pendant 30 minutes renforce l'activation du récepteur Y4 par le polypeptide pancréatique et d'autres peptides (NPY, PYY), tout en accélérant le recrutement de l'arrestine-3[1].

Lignées cellulaires

Cellules COS7 stablement transfectées avec le récepteur Y4 humain-eYEP et la protéine G chimérique △6Gαqi4-myr; cellules HEK293 exprimant transitoirement les protéines de fusion Y4R-Rluc8 et vénus-arrestine-3; muqueuse du côlon descendant murin exprimant endogènement le récepteur Y4.

Méthode de préparation

Pour les essais BRET de recrutement de l'arrestine-3, les cellules HEK293 transfectées ont été ensemencées dans les plaques et incubées avec de la coelentérazine H. Les cellules ont été traitées à tBPC (5.1μM-30μM) pendant 30 minutes.

Conditions de réaction

5.1-30µM; incubation pendant 30 min.

Domaines d'application

tBPC potentialise l'activation du récepteur Y4 par son ligand endogène, le polypeptide pancréatique (PP), ainsi que par d'autres ligands peptidiques (NPY, PYY), à la fois dans les voies qui transmettrent les signaux calciques médiées par les protéines G et de recrutement de l'arrestine-3. tBPC agit comme modulateur allostérique positif (MAP), présente une faible activité agoniste dans les systèmes de surexpression.
