Description of Acyclovir

Acyclovir es un potente medicamento antiviral oral. Acyclovir tiene actividad antiherpética con valores de IC50 de 0.85 μM y 0.86 μM para HSV-1 y HSV-2, respectivamente. Acyclovir induce la interrupción del ciclo celular y apoptosis[1] [2].

En el ensayo de reducción de placas (PRA) en células Vero, el tratamiento con Acyclovir (20 μM; 3 días) mostró una inhibición del 100% de las placas[1]. Acyclovir (10-100 μM; 24-72 horas) bloquea la síntesis de ADN, deteniendo el ciclo celular en las fases G2/M y S, e incrementa el pico hipodiploide G1 de manera dependiente de la dosis. Acyclovir (10-100 μM; 24-72 horas) induce apoptosis a través de la activación de la caspasa-3 y la fragmentación del ADN nuclear[2].

El tratamiento de ratones infectados con Acyclovir (20 mg/kg; vía oral; tres veces al día; 10 días) inhibió el desarrollo de lesiones cutáneas y condujo a una disociación entre la respuesta de hipersensibilidad tardía (DTH) y la producción de anticuerpos[3]. En el modelo de nefrotoxicidad inducida por Acyclovir, después de administrar a los ratones Acyclovir (150, 600 mg/kg; vía intraperitoneal; 9 días), se observaron aumentos en los niveles de creatinina y nitrógeno ureico en suero, cambios histopatológicos en los riñones de los ratones, y se redujeron los niveles de expresión del factor de crecimiento endotelial vascular (VEGF) y su receptor VEGFR2[4].

Método de preparación

Las células Vero fueron infectadas con HSV-1 durante 1 hora y luego tratadas con NPRE (0.1, 0.2, 0.4, 0.6, 0.8 mg/mL). El DMSO se utilizó en las muestras de control y el Acyclovir (20 µM) se usó como control positivo.

Condiciones de reacción

Método de preparación

Ratones ICR fueron divididos aleatoriamente en tres grupos para recibir solución salina, 150 mg/kg de Acyclovir, o 600 mg/kg de Acyclovir una vez al día durante 9 días. Los ratones se mantuvieron a temperatura ambiente y con una humedad del 70±10%, en un ciclo de luz-oscuridad. El agua y la comida estaban disponibles ad libitum. El Acyclovir se administró por vía intraperitoneal (ip) diluido en 20 mL/kg de solución inyectable de cloruro de sodio al 0.9%. El grupo de solución salina recibió un volumen equivalente de la misma solución.

Forma de dosificación

150, 600 mg/kg; ip; 9 días.

Áreas de aplicación

Después de la administración de Acyclovir, los niveles de creatinina sérica y nitrógeno ureico aumentaron, los riñones de los ratones mostraron cambios histopatológicos y una reducción en los niveles de expresión del factor de crecimiento endotelial vascular (VEGF) y su receptor VEGFR2.
