Description of Sphingomab

Sphingomab (LT1002) is a mouse-derived IgG1 κ monoclonal antibody and also an inhibitor targeting S1P, with high affinity for S1P and a K d value of 0.03 ± 0.002 nM. Sphingomab blocks S1P -mediated IL-8 release in ovarian cancer cells. Sphingomab exhibits anti-angiogenic activity in a mouse model of choroidal neovascularization. Sphingomab reduces peripheral blood lymphocyte counts. Sphingomab can be used in the research of ovarian cancer and age-related macular degeneration [1].

In Vivo, Sphingomab (intravitreal injection; administered twice, 1 day before laser treatment and 6 days after laser treatment) inhibits laser-induced choroidal neovascularization by more than 85% in C57Bl/6J mice[1]. Sphingomab (50 mg/kg; intravenous injection; single dose) reduces the peripheral blood lymphocyte count in healthy C57Bl/6J mice to approximately 50% of the baseline level within 24 h[1].

In Vitro, Sphingomab (0.0125-0.4 μg/mL) specifically binds to S1P in a dose-dependent manner[1]. Sphingomab binds S1P with high picomolar affinity, with a Kd of 0.03 ± 0.002 nM as measured by surface plasmon resonance[1]. Sphingomab (2-2500 μg/mL; 1 h) dose-dependently inhibits S1P-mediated interleukin-8 release from human ovarian cancer cell line SKOV3, with an IC50 of 513 ± 57 μg/mL[1].
