In Vitro, Tracer AL244 (6 nM-3 µM; 2 h) generates strong BRET signals in HEK293T cells expressing full-length human TRIM21 with a C-terminally fused NanoLuc, enabling live-cell target binding assays[1]. Tracer AL244 generates strong BRET signals and a favorable assay window when interacting with C-terminally fused NanoLuc-TRIM21, whereas it produces weak signals when interacting with N-terminally fused NanoLuc-TRIM21[1].
