Triptolide ist ein Diterpen-Triepoxid, das erstmals aus der Heilpflanze Tripterygium wilfordii isoliert wurde. Es zeigt immunsuppressive, entzündungshemmende, antiproliferative und antitumorale Wirkungen. Es hemmt die Aktivierung von NF-κB, einer Familie von Transkriptionsfaktor-Proteinkomplexen[1]. Triptolide hat ein breites Spektrum an Antikrebsaktivität und hemmt die Entwicklung von Dickdarmkrebs, Brustkrebs, Nierenzellkarzinom und Gebärmutterhalskrebs[2].
In vitro hemmt Triptolide (50 nM, 72 Stunden) erheblich die Proliferation der Eierstockkrebszellen SKOV3 und A2780. Triptolide (15 nM, 48 Stunden) hemmt die Zellmigration erheblich und blockiert die invasive Fähigkeit der Zellen[2]. Triptolide (50-200 nmol/L), mit PANC-1- und MiaPaCa-2-Zellen inkubiert, reduziert die Zellviabilität erheblich, ohne die Viabilität normaler Pankreasgallengangszellen zu beeinträchtigen[3]. Der IC50-Wert von Triptolide auf die RSF-Zellviabilität ist 74.3 nM und sein IC50-Wert auf die RSF-Zellproliferation ist 20.4 nM[4]. Triptolide (0-150 nM) hemmt erheblich die Viabilität von HK1- FaDu- und C666-1-Zellen dosis- und zeitabhängig[5].
In vivo reduzierte die Behandlung mit Triptolide (1 mg/kg/d, p.o.) bei Nacktmäusen über 3 Wochen die Zahl metastatischer Knoten um etwa 80%[2]. Die Verabreichung von Triptolide (0.2 mg/kg/Tag) an Mäuse mit transplantierten Pankreastumoren über 60 Tage reduzierte das Wachstum von Pankreaskrebs und verringerte die lokale Tumorausbreitug erheblich[3].
