Acyclovir est un puissant médicament antiviral oral. Acyclovir présente une activité anti-hépatique, sa valeur d'IC50 est respectivement 0.85 μM et 0.86 μM pour HSV-1 et HSV-2. Acyclovir induit une perturbation du cycle cellulaire et l'apoptose[1] [2].
Dans l'essai de réduction des plages (RPA) sur les cellules Vero, le traitement avec Acyclovir (20μM; 3j) a montré une inhibition de 100% des plages[1]. Acyclovir (10-100μM; 24-72h) bloque la synthèse de DNA, ainsi donc, entraîne le cycle cellulaire aux phase G2/M et S et augmente le pic hypodiploïde G1 de manière dose-dépendante. Acyclovir (10-100μM; 24-72h) induit l'apoptose via l'activation de la caspase-3 et la présence e fragementation de DNA nucléaire[2].
Le traitement des rats infectés par Acyclovir (20mg/kg; po; trois fois par jour; 10 jours) a inhibé le développement des lésions cutanées et a entraîné une dissociation entre la réponse DTH et la production d'anticorps[3]. Dans le modèle de néphrotoxicité Acyclovir-induite, après administration de Acyclovir (150, 600 mg/kg; ip; 9j), le niveau de créatinine sérique et d'azote uréique est augmenté, des changements histopathologiques sont survenus dans les reins des rats, et le niveau d'expression du facteur de croissance endothéliale vasculaire (VEGF) et de son récepteur VEGFR2 ont été réduits[4].

Méthode de préparation | Les rats ICR ont été aléatoirement divisés en trois groupes pour recevoir la saline, 150mg/kg Acyclovir, ou 600mg/kg Acyclovir une fois par jour pendant 9 jours. Les rats ont été maintenus à la température ambiante et à la humidité de 70±10%, avec un cycle jour-nuit. La nourriture et l'eau étaient disponibles ad libitum. Acyclovir a été administré par voie intra-péritonéale (ip) dilué dans 20mL/kg solution injectable de chlorure de sodium à 0.9%. Le groupe traité avec saline a été administré par un volume égal de la solution injectable de chlorure de sodium à 0.9%. |
