Gap 26は、コネキシン模倣体ペプチドとして、コネクソンのドッキングに決定的に寄与し、ギャップジャンクションを生成するSHVRアミノ酸領域を持つ[1]。Gap 26は、カルシウムの増加に誘発されたATPの放出を阻害できる[2]。Gap 26は、ギャップ結合を媒介した細胞ペアにおける電気的な結合を阻害した[3]。
Gap 26 (25μM；24時間)は尿細管上皮細胞のアポトーシスを弱めた[4]。Gap 26 (150μM；24時間)はラットの肺胞II型上皮細胞(RLE-6TN)で、ROS生産を減らし、ASK1-JNK/p38シグナル伝達を阻害し、アポトーシスを減らした[5]。
Gap 26 (1μg/kg/日；腹腔内；8週間、週に3回)は肝硬変性心筋症ラットモデルで、変時性低反応性と肝臓損傷の程度を弱め、心臓に抗酸化作用を持つ[6]。Gap 26 (25μg/kg；腹腔内；4日間)は中大脳動脈閉塞症(MCAO)ラットモデルで、シナプスの破壊を更に悪化させ、海馬の脊柱密度を低くした[7]。

準備方法 | ラットを10%の抱水クロラールで麻酔した。その後、ラットの首に正中切開を行った。右総頸動脈(CCA)、外頸動脈(ECA)と内頸動脈(ICA)を分離した。ECAを総頸動脈分岐部の約4mm上で切断した。抵抗性が出るまで、断端から4.0-sのナイロンモノフィランメントをICAに挿入した。こうして、中大脳動脈（MCA）への血流は遮断された。2時間後、フィラメントを再潅流のため取り出し、切開された部位を結紮した。Gap 26を生理食塩水に溶解し、25μg/kgの用量で毎日一回腹腔内投与した。急性期における影響を避けるため、虚血後の3日目（梗塞量が安定し、回復期が始まる）に、Gap 26を応用することにした。MCAOの7日目に、ラットを殺処分し、脳をできるだけ早く除去した。快速ゴルジ染色キットを、組織の調製と染色手続きのため、応用した。総ゴルジ-Cox染色手続きを、指示通りに厳密に実行した。冷凍ミクロトームを使い、ブレグマから+1.00から5.00mmで始め、各ブロックを冠状方向に厚さ100μmの連続切片に切った。切片をスライドの上に置き、レーザースキャン共焦点顕微鏡で観察した。 |
