Preparation Method

HCAECs were exposed to Angiotensin II (0, 0.1, 1, 5, 10, 20, 50, and 100 nmol/L) for 24 hours.

Reaction Conditions

Angiotensin II (0, 0.1, 1, 5, 10, 20, 50, and 100 nmol/L) for 24 hours

Applications

Angiotensin II (1 nM) induces the expression of LOX-1 and VEGF and enhances capillary formation from human coronary endothelial cells in Matrigel assay. Angiotensin II -mediated expression of LOX-1 and VEGF, capillary formation, intracellular reactive oxygen species generation, and phosphorylation of p38 as well as p44/42 mitogen-activated protein kinases, are suppressed by anti-LOX-1 antibody, nicotinamide-adenine dinucleotide phosphate oxidase inhibitor apocynin and the Angiotensin II type 1 receptor blocker Losartan, but not by the Ang II type 2 receptor blocker PD123319.

Preparation Method

To distinguish the AT1 receptor population that is critical for the pathogenesis of hypertension, osmotic minipumps were implanted s.c. into each animal to infuse Angiotensin II (1,000 ng/kg/min) continuously for 4 weeks.

Dosage form

Angiotensin II (1,000 ng/kg/min) for 4 weeks.

Applications

AT1 receptors in the kidney are primarily responsible for the actions of Angiotensin II to cause hypertension.

안지오텐신 II는 혈관 수축제이자 레닌/안지오텐신 시스템의 주요 생체 활성 펩타이드입니다. 안지오텐신 II는 인간의 혈압 조절에서 중심적인 역할을 하며, 주로 안지오텐신 II 및 G 단백질 결합 수용체(GPCRs) 안지오텐신 II 유형 1 수용체(AT1R)와 안지오텐신 II 유형 2 수용체(AT2R)가 상호작용하여 매개합니다.[1]

인간은 LOX-1 종속적인 산화환원 민감한 경로를 통해 내피세포에서 모세혈관 형성을 유도합니다. 안지오텐신 II(1 nM)는 LOX-1과 VEGF의 발현을 유도하고, 인체 심장 내막내 세포에서 Matrigel 실험에서 모세혈관 형성을 촉진시킵니다[3,4]. 안지오텐신 II는 염분과 물의 총 신장 관류를 조절하는데 도움이 되며, AT1 수용체 종속적 기전을 통해 상피 세포 나트륨 채널 활동을 직접 자극합니다[8]. 안지오텐신 II는 혈관 평활근 세포의 성장을 유도하고, 파이브로브라스트에서 콜라겐 I 및 III 합성량을 증가시켜 혈관벽 및 심근의 두께를 늘리며, 섬유증을 유발합니다. 안지오텐신 II는 아파치스도 유발합니다[2]. 안지오텐신 II가 혈압 상승에 영향을 주는 것은 AT1 수용체를 매개로 합니다[1]. 이러한 수용체는 심장, 신장, 혈관, 부신선 및 뇌의 순환 조절 센터와 같은 혈압 조절에 중요한 역할을 하는 다양한 기관계에서 발현됩니다[5]. 뇌에서 안지오텐신 II의 빈뇨 주입은 AT1A 수용체 매개로 극적인 압력 반응을 유발합니다[6].

쥐에서 신장 내 AT1 수용체는 Angiotensin II의 고혈압 유발 작용을 주로 담당합니다. vitro 상황에서 ROS/펩티딜 아르기닌 디미나아제 유형 4 및 자가포식 종속적 경로를 통해 Angiotensin II 유도 NETosis. Angiotensin II 수용체 차단제로 시작된 EH 환자에서 순환 NET 및 혈전 생성 농도가 크게 감소하였으며, 그들의 혈장은 프로응고성 NETs를 유발하지 못하였습니다.
