Description of SIS3 HCl

SIS3 HCl은 Smad3 억제제로 IC50 3 μM로 Smad3의 인산화를 억제한다.

SIS3 HCl은 농도에 따라 증가된 p3TP-lux 루시퍼아제 활동을 제거하고 변화 성장 인자(TGF-1)에 의해 유도된 Smad3의 인산화와 Smad3과 Smad4 간의 상호작용을 완화시킬 수 있고 SIS3 HCl은 Smad2의 인산화에 영향을 미치지 않는다. SIS3 HCl은 전사활성성을 감소시키는 방식으로 TGF-1의 효과를 완화시키고 TGF-1을 통해 성체 근섬유아세포의 분화를 억제한다 [1]. ML-12 세포에서 10 μM의 SIS3 HCl은 TGF-β를 통한 Smad3의 인산화를 차단했고 Smad2/3의 총량은 변하지 않았다 [2].

Tie2-Cre; Loxp-EGFP 쥐 연구에서 SIS3 HCl(2.5 mg/kg/일)은 스트레포토시신(STZ) 유발 당뇨병성 신증에서 Smad3 활성을 억제했다. SIS3 HCl은 엔도테리얼 메센치멀 전환을 제거하고, 신장섬유증을 줄이고 신장질환의 진행을 늦추었다[3]. 고지식 식이로 유도된 2형 다이어벳(T2DM) 모델 쥐에서 SIS3 HCl(5 mg/kg)은 체중, 지방량, 그리고 빵먹은 혈당을 현저하게 낮추었고 쥐의 인슐린 민감도와 구강 내당능도 개선했다[4].
