NT157はインスリン受容体基質-1/-2(IRS-1/2)の選択的な阻害剤である[1]。IRS-1/2は重要な細胞質アダプタータンパク質であり、インスリンとインスリン様の成長因子1(IGF-1)のシグナル伝達を介し、糖代謝、細胞の増殖と発生で肝心な役割を果たしている[2]。NT157は、アポトーシスを誘発し、肝心なシグナル伝達経路を破壊することでその作用を発揮し、様々な疾患(メラノーマ、前立腺癌や大腸癌など)の薬剤研究と開発によく使われている[3,4]。
In vitro(体外)実験で、骨肉腫MG-63とU-2OS細胞をNT157(3μM)で24時間前処理したところ、細胞の遊走能力が著しく阻害され、同時にIRS-2タンパク質の発現が低下した[5]。H1299とH460細胞をNT157(3.2、6.4、12.5μM)で7日間処理したところ、コロニー形成能力が用量依存的に著しく阻害された;12.5μMの濃度で、H1299とH460細胞のコロニー形成率はそれぞれ対照群の1.4%と0.8%に減少した[6]。
In vivo(体内)実験で、SMMC-7721細胞の尾静脈注射により作製された肝細胞癌の肺転移のヌードマウスモデルに、NT157(100mg/kg;毎日二回)を4週間腹腔内注射したところ、対照群と比べ、肺転移の発生率が著しく低下した[7]。A375SM細胞を皮下接種したマウスに、NT157(70mg/kg;週に三回)を4週間静脈内注射し、最終投与の48時間後に、腫瘍組織におけるpY(705)Stat3のレベルは著しく低下した[4]。

調製方法 |
1×103個の細胞/ウェルの密度で、H1299とH460細胞を6ウェルのプレートに播種し、24時間インキュベートし、その後NT157(3.2、6.4と12.5μM)で7日間処理した。コロニーを固定し、1%のクリスタルバイオレット溶液で染色し、コロニーの数を数えて、コロニー形成能を評価した。 |
