ClAP (Alkaline Phosphatase, Calf intestinal) is a membrane-bound glycoprotein that hydrolyzes a broad range of monophosphate esters at alkaline pH [1]. ClAP can detoxify various inflammatory mediators such as lipopolysaccharide (LPS), lipoteichoic acid, CpG DNA, and adenosine triphosphate [2]. ClAP has been widely used to alleviate the LPS toxicity in mice and piglets, and to enhance the survival of mice after Escherichia coli infection[3].
In vivo, ClAP treatment via oral administration at a dose of 700U/kg daily for 6 days significantly improved experimental colitis in rats and reduced bacterial translocation[4]. 15 minutes after the cecal ligation and perforation surgery, a single intravenous injection of ClAP (0.15IU/g) can alleviate the inflammatory response of secondary peritonitis in mice and reduce the associated liver and lung damage[5].

Preparation Method |
Female Wistar rats (175-225g) were housed in cages and maintained in air-conditioned animal quarters with a 12h light–dark cycle and free access to autoclaved tap water and a standard chow diet. rats were fasted overnight and anesthetized with halothane. Under these conditions, rats were given 10mg of trinitrobenzene sulfonic acid (TNBS) dissolved in 0.25ml of 50% ethanol (v/v) by means of a cannula inserted 8cm through the anus. Rats were kept in a head-down position for an additional 30s and returned to their cage. Rats were administered 700U/kg/day of ClAP by the oral route. Oral treatments started 1h before colitis induction. Afterward treatments were given daily until mice were sacrificed (cervical dislocation) at day 6. Food and water intake and body weight were determined daily. Colon assessment and analysis of bacterial translocation. |
