ML089 is an orally active and membrane-permeable phosphomannose isomerase (PMI) inhibitor (IC₅₀=1.3µM) that blocks the conversion of mannose-6-phosphate to fructose-6-phosphate. ML089 can be used in studies related to type Ia congenital disorder of glycosylation (CDG‑Ia)[1-3].
In vitro, HeLa cells were preincubated with ML089 (20–40µM) for 2h, after which [³H]-mannose and [³⁵S]-Met/Cys were added and the cells were incubated for an additional 1h. ML089 elevated [³H]-mannose incorporation into precipitated (TCA-precipitated) proteins, accompanied by a reduction in [³⁵S]-Met/Cys incorporation[2]. Mouse embryonic stem cell-derived gastruloids were treated with ML089 (30µM) from day 3 to day 4. ML089 reduced the overall gastruloid volume and exerted a mild effect on axial elongation[3].
In vivo, C57BL/6 mice were administered ML089 at 5mg/kg via single intravenous injection and at 20mg/kg via single oral gavage. ML089 yielded quantifiable plasma exposure, with a post-dose Cmax of approximately 162nM, Tmax of approximately 45min, AUC₀₋ₜ of approximately 305nM·h, and t₁/₂ of approximately 1.3h, and the oral bioavailability reached 35%[2].

Preparation Method | HeLa cells were seeded and grown to approximately 70% confluency in 24-well plates. Cells were incubated with ML089 for 2h at 37°C, followed by simultaneous addition of 50μCi/mL [2-³H]-mannose and 5μCi/mL ³⁵S-Met/Cys Trans label and further incubation at 37°C for 1h. |

Applications | ML089 produced a dose-dependent increase in ³H-mannose incorporation normalized to ³⁵S-Met/Cys incorporation in HeLa cells, indicating diverted mannose flux from catabolism toward glycoprotein N-linked glycosylation via inhibition of cellular phosphomannose isomerase (PMI). Over the tested concentration range, ML089 showed enhanced glycosylation signal with a favorable cellular efficacy–toxicity profile; decreased ³⁵S-Met/Cys incorporation at higher concentrations was observed but was attributed by the authors to off‑target activity rather than to on‑target PMM2 inhibition, as PMM2-deficient CDG-Ia patient fibroblasts grew normally in the presence of the PMI inhibitors without reduced ³⁵S-incorporation. |
