Zeatin is a naturally occurring cytokinin-like plant hormone [1]. Zeatin binds to cytokinin receptors, activating downstream signaling pathways, promoting cell division, differentiation, and chloroplast development, and delaying leaf senescence [2-3]. Zeatin is commonly used in anti-aging and cell regulation [4].
In tobacco, Zeatin (10μM; 24h) inhibits symptom development of Pst infection [5].
In red blood cells, Zeatin (40-200μM; 24h) causes significant hemolysis and leakage of K+, Na+, AST, and LDH [6]. In human dermal fibroblasts, Zeatin (1-200μM; 48h) treatment prevented cell enlargement, reduced intracellular debris, prevented actin polymerization, and enhanced the cells ability to break down hydrogen peroxide and respond to ethanol and oxidative stress [7].
In male ICR mice, Zeatin (1.5-4.5mg/kg; po; 4 weeks) treatment significantly improved scopolamine-induced amnesia [8]. In rat depression model, Zeatin (10mg/kg; ip; single injection) treatment produced antidepressant-like effects [9].

Preparation Method | To address the effects of zeatin on plant immunity, an experimental design using zeatin in the tobacco-Pst pathosystem was employed. Four detached leaves from 7–10-week-old tobacco SR1 plants grown under greenhouse conditions were fed through the petiole with 10μM zeatin diluted from a 100mM stock solution in 0.5M NaOH; correspondingly diluted NaOH solutions served as solvent controls 24 hours before Pst infection. |

Preparation Method | Zeatin (40-200μM) was dissolved in DMSO and added to a healthy donor red blood cell suspension. The cells were incubated at 37℃ for 24h in different Ringer buffers. Hemolysis was then measured (absorbance at 405nm, with ddH2O as 100% control), and hemolysis markers such as AST, LDH, K+, and Na+ were measured using a clinical biochemistry analyzer. |

Animal models | Male ICR mice |
Preparation Method | Male ICR mice, weighing 25–30g, were used for passive avoidance testing following a 1-week adaptation period (20–23℃; 12-hour light cycle from 09:00 to 21:00; food and water ad libitum). Mice were allowed free access to normal feed or feed mixed with zeatin for 4 weeks. Amnesia was induced in mice with scopolamine (1mg/kg of body weight) given subcutaneously. Behavioral tests were started 30 minutes after injection. |
