Description of H-Gln-AMC

H-Gln-AMC es un sustrato fluorogénico utilizado para medir la actividad de las enzimas glutamil ciclasa (QC) con un valor de Km de 51 ± 3µM a pH 8.0 y un valor de kcat de 5.4 ± 0.1s-1[1]. La glutaminil ciclasa (QC) cataliza la ciclización intrmolecular de los residuos de glutaminilo N-terminal glutaminyl (Gln) y glutamyl (Glu) para formar péptidos o proteínas pyroglutamic (pGlu). Algunos hallazgos recientes sugieren que el control de calidad puede ser un objetivo potencial contra la enfermedad de Alzheimer, la enfermedad de Huntington y otras enfermedades neurodegenrativas graves[2].

H-Gln-AMC se utilizó como sustrato fluorescente para detectar la actividad de la glutaminil ciclasa (QC) en el líquido cerebrospinal (CSF) de pacientes con esclerosis múltiple (MS).En el CSF, H-Gln-AMC fue catalizado por QC para formar pyroglutamyl-AMC, que posteriormente fue hidrolizado por piroglutamil aminopeptidasa para liberar el producto fluorescente AMC. Los resultado muestraran un ligero aumento en la actividad de QC observado en pacientes con MS, aunque la diferencia no fue significativa[3]. H-Gln-AMC se utilizó como sustrato fluorescente para medir la actividad de las glutaminil ciclasas (QCs) de Porphyromonas gingivalis (PgQC), Tannerella forsythia (TfQC), y Prevotella intermedia (PiQC). A una concentración de 50μM H-Gln-AMC, la actividad catalítica de PgQC, TfQC y PiQC podría determinarse con precisión en función de los cambios en la intensidad de fluorescencia de AMC[4].

Protocol of H-Gln-AMC

H-Gln-AMC utilizdo como sustrato para ensayos de actividad de control de calidad.

1. Prepare un tampón Tris-HCl de 50mM con NaCl de 150mM a pH 8.0.

2. Prepare soluciones de H-Gln-AMC con concentraciones variables (e.g 50µM para pruebas de actividad catalítica de control de calidad bacteriana) en el tampón Tris-HCl.

3. Agregue 0.5U de pyroglutaminyl aminopeptidase a cada pozo de una placa de microtiter de 96 pozos que contiene las soluciones H-Gln-AMC.

4. Incubar el plato a 30°C durante 10 minutos.

5. Agregue aproximadamente 50nM de la enzima QC a un volumen final de 125μL de mezcla de reacción para iniciar la reacción.

6. Monitoree la intensidad de fluorescencia a longitudes de onda de excitación/emisión de 380/460nm usando el FluoStar Optima

7. Calcule la actividad de control de calidad a partir de una curva estándar del fluoróforo AMC generado bajo las mismas condiciones de ensayo
