Description of Acevaltrate

Acevaltrate is an active component extracted from the roots of Valeriana glechomifolia, which inhibits Na+/K+-ATPase in the rat kidney and brain hemispheres with IC50s of 22.8μM and 42.3μM[1, 2]. Acevaltrate has antitumor effect on various cancer types, and can induce cell apoptosis and inhibit proliferation through the HIF-1α, Otub1/c-Maf, and USP10/CCND1 axes[3, 4]. Acevaltrate can inhibit A549, PC-3M, HCT-8, and Bel7402 cell lines with IC50 values of 2.9µM, 1.4µM, 1.0µM, and 1.7µM, respectively[5].

In vitro, Acevaltrate (0, 2 and 4μM; 24h) downregulated CCND1 protein expression in glioblastoma cells in a concentration-dependent manner[4]. Acevaltrate (0, 1, 2 and 5μM; 1h) significantly downregulated PCBP1 and PCBP2 proteins in a dose-dependent manner in RKO cells[6].

In vivo, Acevaltrate (0, 10, 25, and 50mg/kg; 22d; p.o.) suppressed tumor growth in nude mice inoculated with RKO cells[6].

Protocol of Acevaltrate

Cell experiment [1]:

Cell lines

Glioblastoma cell lines (U251)

Preparation Method

Cells were seeded in 96-well plates and treated with Acevaltrate for 24h. Then the cells were subjected to Annexin V/PI staining and flow cytometric analyses, and the cell lysates were subjected to immunoblot assays against PARP, Caspase-3 and pro-survival proteins.

Reaction Conditions

0, 2 and 4μM; 24h

Applications

Acevaltrate induced 40% cell death at 4μM, caused PARP and Caspase-3 cleavage, and downregulated the expression of Bcl-2 and Mcl-1 in a dose-dependent manner.

Animal experiment [2]:

Animal models

Male nude mice

Preparation Method

Mice were subcutaneously inoculated with RKO cells (5×106 cells). After subcutaneous inoculation of tumor cells, mice were randomly grouped according to tumor volume and body weight when the tumor volume reached 50mm3. Acevaltrate was orally administered at doses of 0, 10, 25, and 50mg/kg once daily for 22 days (corn oil as a control). Tumor size intensity was measured every 2-3days.

Dosage form

0, 10, 25 and 50mg/kg; 22d; p.o.

Applications

Acevaltrate treatment at 10, 25 and 50mg/kg significantly reduced tumor size, volume, and weight compared to the control group.
