Description of AK59

AK59 actúa como degradador de la proteína STING y se apoya en la ligasa E3 HERC4 con HECT-domain para cumplir su función[1]. Este compuesto acelera la degradación de STING vía el sistema ubiquitina-proteasoma, con lo que logra bloquear la vía de señalización cGAS/STING[2]. En ensayos bioquímicos suele emplearse AK59, ideal para filtrar inhibidores de STING y agentes degradantes dirigidos exclusivamente a variantes mutadas de STING[3].

Células THP1

Método de preparación

Las células THP1 se cultivaron en un medio RPMI 1640 que contenía un 10% de suero bovino fetal (FBS), 2mM de l-glutamine, 25mM de Hepes, 1mM de sodium pyruvate y 50U/ml de penicillin y 100mg/ml de streptomycin a 37℃ en presencia de 5% de CO2. 2×106 por pozo en una placa de seis pocillos se sembraron y trataron con diferentes concentraciones de AK59 (1, 2, 5, 7.5, 10µM) durante 16h. Posteriormente, se realizó un análisis de Western blot.

El tratamiento con AK59 redujo la expresión de la proteína STING en las células THP1 de manera dependiente de la dosis..
