AK59 est un dégradant de STING qui utilise HERC4, une ligase E3 à domaine HECT[1]. AK59 inhibe les voies qui transmettrent les signaux de cGAS/STING en favorisant la dégradation de STING par le système ubiquitine-protéasome (UPS)[2]. AK59 est fréquemment utilisé dans les expériences biochimiques à cibler les inhibiteurs de STING et les dégradants spécifiques aux mutants de STING[3].

Méthode de préparation | Les cellules THP1 ont été cultivées dans un milieu RPMI 1640 contenant 10% de sérum de veau foetal (FBS), 2mM de L-glutamine, 25mM de HEPES, 1mM de pyruvate de sodium, et 50U/ml de pénicilline et 100mg/ml de streptomycine à 37°C en présence de 5% CO2. 2×106 cellules par puits dans une plaque à six puits ont été semées et traitées avec différentes concentrations d'AK59 (1, 2, 5, 7.5, 10µM) pendant 16h. Ensuite, une analyse Western Blot a été réalisée. |
