Beta-Sitosterol es un esterol vegetal con múltiples actividades biológicas, incluyendo actividades antiinflamatorias, antitumorales, antipiréticas e inmunomoduladoras[1, 2]. Beta-Sitosterol puede inhibir la migración de las células musculares lisas vasculares a través de la vía PPARG/AMPK/mTOR[3].
In vitro, el tratamiento con Beta-Sitosterol (10-160μM) de las células MH7A durante 12-48h inhibió la proliferación celular de una manera dependiente de la concentración y el tiempo, redujo significativamente la expresión de los genes MMP9, HSP90AA1, SRC, EGFR y ALB, y aumentó significativamente la expresión de la proteína CASP3[4]. Tratamiento con Beta-Sitosterol (0.2-1mM) de células de cáncer de hígado humano HepG2 durante 24h indujo la citotoxicidad de una manera dependiente de la dosis e indujo la acumulación intracelular de ROS[5].
In vivo, Beta-Sitosterol (50, 100, 200mg/kg) se inyectó por vía intraperitoneal en ratas con edema en la pata y edema inhibido de manera dependiente de la dosis, con el efecto inhibidor que alcanzó el 50-70%[6].

Método de preparación | Utilizó 30 ratas organizadas en 6 grupos con 5 individuos cada uno: un grupo de control negativo que se administró intragástrico (IG) con aceite mineral; otros tres grupos que fueron IG administrados con Beta-Sitosterol (50, 100 y 200mg/kg), previamente diluidos en aceite mineral; y finalmente, dos grupos más intraperitonealmente (i.p.) administrados con medicamentos antiinflamatorios conocidos, en los que a un grupo se le inyectó 200mg/kg de ibuprofeno (un agente antiinflamatorio no esterioideo), y al otro grupo se le inyectó 10mg/kg de prednisona (un agente antiinflamatorio esteroidal). A todos los animales se les inyectó inmediatamente 0.1mL de anticuerpo anti-ovalbumin (diluido 1:3) en el cuatro trasero del área plantar de la pierna derecha y en la pierna contralateral; a las ratas se les inyectó 0.1mL de NaCl al 0.9% . En un paso posterior, todos los animales fueron inyectados por vía intravenosa (i.v.) con una suspensión de albúmina de huevo (25mg/kg). El volumen de la pierna hinchada se determinó con un pletismómetro digital 3 horas después de la inyección de ovoalbúmina. El valor del edema de la pata se estableció determinando la diferencia entre el volumen obtenido en la pierna tratada y en el control (oierna izquierda). Para el cálculo de los datos, se consideró que el volumen de la pierna hinchada representaba el 100% de la inflamación. |
