Physostigmine es un inhibidor reversible de la acetylcholinesterase (AChE) que atraviesa las membranas celulares, penetra en la barrera hematoencefálica y estimula la neurotransmisión colinérgica central[1,2].Acetylcholinesterase se encarga de degradar la acetilcolina utilizada. Al interferir con metabolismo de acetilcolina, la fisostigmina estimula los receptores nicotínicos y muscarínicos, lo que provoca un elevamiento de la acetilcolina disponible en sinapsis. La fisostigmina se utiliza en la investigación sobre vías de señalización colinérgica y modelos de trastornos vinculados, así como en el tratamiento del glaucoma y la toxicidad anticolinérgica[3, 4].
In vitro, Physostigmine (0.2-2μM) se incubó con eritrocitos AChE de humanos, mono erhesus, cerdos y conejillos de indias, y la curva de inhibición enzimática se monitoreó durante 30 minutos con el fin de calcular la constante cinética de inhibición bimolecular (ki), y los valores constantes de inhibión mostraron una variación mínima entre las especies (Humanos: 5.40 × 106M-1 min-1; mono Rhesus: 6.34 × 106M-1 min-1; Cerdo: 4.14 × 106M-1 min-1; Conejillo de Indias: 3.50 × 106M-1 min-1 )[5]. La incubación def Physostigmine (1mM) con suero de humanos con función hepática normal mientras 120 minutos inhibió el procesos de desacilación de grelina acilada (AG) en el suero, lo que resultó en un nivel residual más alto de AG después de la incubación en comparación con grupo control[6].
In vivo, la administración inmendiata de Physostigmine (0.1mg/kg) a través de inyección intraperitoneal a ratones después de la exposición hipóxica (FiO2 = 0.1) mejoró el rendimiento de memoria breve de los ratones en novedosa prueba de reconocimiento de objetos, previniendo el deterioro cognitivo inducido por hipoxia[7]. La inyección subcutánea de Physostigmine (0.4mg/kg) administrada 15 minutos antes de las pruebas de comportamiento en ratones C57BL/6J redujo la amplitud máxima de sobrestredo en respuesta a un estímulo acústico de 120dB y prolongó el tiempo para alcanzar la amplitud máxima[8]. El tratamiento de un modelo de ratón de sepsis inducido por ligadura y punción cecal (CLP)- con Physostigmine (80μg/kg; 3 veces/díay; 3 díays; i.p.) mejoró la proporción de supervivencia de los ratones[9].
